Development and Evaluation of Sugarcane Somaclones

Abstract/ Executive Summary:

This experiment was conducted to develop red rot, salt and drought tolerance in sugarcane varieties or clones at BSRI farm, Ishurdi, Pabna during 2009-2010 cropping season. The experiment comprises three varieties viz. Isd 32, Isd 35 and Isd 36. The experiment was laid out in randomized complete block design (RCBD) with three replications using plot size of 5 m × 4 m. Planting materials showed significant variation on tiller number, millable cane and cane yield. The highest tiller production (171.22) and millable cane (131.60) were found in tissue culture plants. The tissue culture plants gave significantly highest cane yield (73.14 t ha-1) than other planting materials and it was approximately 1.4 times higher than three budded setts (52.56 t ha-1). The budchip settlings produced lowest cane yield (38.09 t ha-1), lowest number of tiller (67.16) and millable cane (56.22). The pol % cane was not significantly affected by the sources of planting materials. The highest pol % cane (12.55) was obtained from budchip settlings and lowest (11.72) was obtained from three budded setts planting.

Objectives:

a. To study the effects of red rot toxin, NaCl, hydroxyl prolin and polyethylene glycol concentrations on different in vitro stages. b. To study the effects of red rot, salinity and drought on regenerated plants under ex vitro conditions. c. To develop red rot, salt and drought tolerance in sugarcane varieties or clones.

Methodology:

a. BSRI released varieties prone to biotic and abiotic stress were used as experimental materials. b. Somaclones from defective varieties were produced through callus culture. c. For callus initiation explants were inoculated on MS + 2,4-D(3mg/l) + 10% coconut water. d. Callus were sub-cultured on the medium containing red rot fungal toxin isolated using standard procedure (5.0, 7.5, 10.0, 12.0, and 15.0%), NaCl (50, 100, 150, 200 and 250 mM), HYP (0.5, 1.0, 1.5, 2.0 and 2.5 mM) and PEG (5.0, 7.5, 10.0, 12.5 and 15.0%). On every concentration the callus were maintained for 2-3 weeks. e. The tolerant callus were tested for retention of tolerance by sub-culture first on media without selecting agents, followed by selecting agents containing media. f. Shoot were regenerated on shooting media (MS+2mg/l BAP + 1mg/l Kn) with and without selecting agents. g. For root initiation regenerated shoots were sub-cultured on the medium MS + NAA (5mg/l) with and without selecting agents. h. After hardening, plants were tested under induced red rot, salinity and drought condotions under pot and field conditions.
