Abstract/ Executive Summary:

The experiment was conducted during the year 2000-2001 at Bangladesh Jute Research Institute Dhaka, Jute Waste Processing Department to Compare between enzymatic and chemical designing. The medium composition and the concentrations of the ingredients were optimized for the production of alpha-amylase by Bacillus amyloliquefaciens in shake flasks using Graeco-Latin Square design technique.The optimized medium composition for alpha-amylase production was found to be 0.85% corn flour,0.5%peptone,0.3% Na-nitrated and 0.79%Potassium dihydrogen phosphate and Di-potassium hydrogen phosphate.The effect of initial PH, incubation temperature and the incubation period for alpha-amylase production B.amyloliquefaciens were also determined. The organisms produced highest level of enzyme in shake-flask culture. The enzyme yield was increased about six-folds as compared to the initial shake flask culture.

Objectives:

To replace chemical desizing for improving the quality of bleaching, dyeing, printing and finishing ensuring the pollution free industrial environment.

Methodology:

Bacillus amyloliquefaciens, a wild bacterial strain isolated in this laboratory identified at the Deutsche Sammlung von Mikroorganismen and Zellkulturen (DMS), Germany was taken for the project. Media used: Two types of media were used. These are:i)Seed culture medium & ii)Fermentation medium Seed culture medium: The composition of seed culture medium was almost same as the enriched medium, but it lacked soluble starch and agar. ii)Fermentation medium:The medium contains the following ingredients:-Potassium dihydrogen phosphate-0.3%,Di-potassium hydrogen phosphate-0.6%, Di-ammonium sulphate-0.1%, Yeast extract-0.2%, Trace element-1%. a)Cells were separated by centrifugation and the supernatant was collected to determined extracellular alpha-amylase activity. b)The amount of reducing sugar was calculated from the standard curve. c)Soluble protein in the supernatant, amylase activity were determined. Optimization of Fermentation medium:Graeco-Latin square technique was used to select the type and concentration of potential carbon,organic & inorganic nitrogen sources and buffering agent. Optimization of culture conditions: The influence of initial PH in the cultures on the production of alpha-amylase was investigation using the optimized medium in shake-flask cultivation of B.amyloliquefaciens.

Achievement/Findings:

Fermentation medium for alpha-amylase production has been optimized where corn flour, rice flour and rice husk as carbon source gave better results. Carbon source, its response to reaction PH and temperature.Further investigation concerning purification of the extracellular alpha-amylase and its kinetic studies are being carried out.After completion of these experiments with better results one can go for enzyme production in large quantities using Bio reactors.
