Effect of Gamma Radiation on Different Aged Pupae of Jute Hairy Caterpillar, Spilarctia Obliqua (walker)

Abstract/ Executive Summary:

Effects of gamma radiation in ten dose viz. 1, 3, 5, 10, 20, 30, 40, 50, 75 and 100 krad were studied on different ages pupae of Spilarctia obliqua. Radiosensitivity of pupae was found to be decreased as age increased. More than 50 per cent adult emergence reduced at 75 krad for 6 days old pupae and 40 krad for 4 and 5 days old pupae. The 10 krad radiation dose prevented 95-100% adult emergence when exposed at younger (1-3 days old) pupae. Deformity rate of emergence adult were greater when the pupae were treated at higher dose. Lowest (11.76 and 17.47%) deformed adults were found from 4 days old pupae at lowest (1 aM 3 krad) dose. The sex ratio of emergence adult from the different doses irradiated pupae resulted more female emerged than that of male. The females of S. oblique from low dose (1-20 krad) irradiated pupae had little effect of egg deposition in the ovary as compared to those of control. The egg laying capacity of female from young (1-3 days old) pupae were highly affected by the different doses radiation. The oldest (4-6 days old) pupae at 1-10 krad had least effect but 20 krad or above the number of eggs laid were drastically reduced.

Objectives:

To examine the response of various ages (1-6 days old) pupae of the S. obliqua to 1, 3, 5, 10, 20, 30, 40, 50, 75 and 100 krad of gamma radiation.

Methodology:

Jute hairy caterpillar were reared at 28.0 ± 0.5°C and 78.5 ± 5 R. H. with 12-hr. photoperiod using jute leaves and rearing techniques described by Islam et at. (1998). In preparation for pupal exposure, large number of larvae were reared. Irradiation were produced by a 1500 Curie Cobalt 60 source. A dose rate of about 0.58 Gy/min was used for all treatment. One, two, three, four, five and six days old pupae were irradiated with doses of 1, 3, 5, 10, 20, 30, 40, 50, 75 ad 100 krad. The pupae were placed in the test-tube during irradiation. Untreated pupae were used as control. Five replicates were taken for each treatment using five pupae per replicate. After irradiation the treated and untreated (control) pupae were held in beaker until adult emergence. Number of emerged normal adult moths with deformed wings were recorded as well as the emerging male and female moths was counted. The number of eggs laid by the females from different doses irradiated pupae were recorded. The females from irradiated and non irradiated pupae were dissected after death and examined and counted the number of eggs deposited in the ovary. The 10 females were dissected for each dose and control.

Achievement/Findings:

Results of these tests demonstrated that the irradiation dose of 1-10 krad is enough to render the adults of jute hairy caterpillar sterile. However, the data clearly showed that the treatment should be limited to older pupae since the sensitivity of pupae of S. obliqua to gamma radiation decreased as they became older. This is one of the main criteria on which any future programme of Sterile Insect Technique (SIT) against jute hairy caterpillar may be based.
