In Vitro Propagation of Stevia (stevia Rebaudiana Bert.)- an Elite Sweetening Herb

Abstract/ Executive Summary:

The experiment was conducted at Biotechnology Laboratory, Bangladesh Sugarcane Research Institute (BSRI), Ishurdi, Pabna, Bangladesh. Shoot tip, nodal segment and leaf base with petiole of Stevia were cultured on MS media supplemented with varying concentrations of BA for shoot production. Nodal segment initiated shoots earlier than shoot tip explant. Higher number of shoot per culture and micro-cuttings were obtained from shoot tip explant than nodal segment. Leaf base with petiole failed to produce shoot on any of the media used. The highest number of shoots (3.2) and micro-cuttings (5.8) were found in the MS medium with 3.0 mgl^-1BA and 2.0 mgl^-1BA respectively. Interaction effects of explants and shooting media were significant for days of shoot initiation, number of shoots per culture and number of micro-cuttings per culture. Among four media the highest number of root per shoot was obtained in MS medium supplemented with 1.0 mgl^-1 NAA followed by 1.5 and 0.5 mgl^-1 NAA respectively. No roots were produced in the medium without auxin (NAA).

Objectives:

To determine plant, dairy products, or juices sweeten by in vitro multiplication of Stevia.

Methodology:

The experiment was conducted at Biotechnology laboratory, Bangladesh Sugarcane Research Institute (BSRI), Ishurdi, Pabna, Bangladesh. The explants, shoot tips, leaf base with petiole and nodal segments were collected from 4-6 month old yard grown plants at Biotechnology Laboratory, BSRI. The explants were cut into small pieces (about 1 cm long) and then were treated with 1% savlon for 5-6 min with constant shaking and washed thoroughly with distilled water. Then the explants were taken under laminar airflow cabinet and surface sterilization was done with a 0.1 % mercuric chloride solution containing two to three drops of tween-80 for five min under aseptic condition and then washed four times with sterilized distilled water. After steriliztion explants were inoculated aseptically for in vitro propagation on MS medium with different concentrations and combinations of growth regulators. For shoot induction explants were inoculated on media MS supplemented BA (1.0, 2.0 and 3.0 mql^-1). Shoots were rooted on the media MS supplemented with NAA (0.5, 1.0 and 1.5 mgl^-1). Visual observation of culture was made every week. Data were recorded after 2 weeks of culture and subjected to statistical analysis using computer software MSTAT- C. Means were compared using Dancan's Multiple Range Test (DMRT).

Achievement/Findings:

Results indicate that shoot tip and nodal segment initiated growth of shoots on hormone free medium within shorter period of time than hormone supplemented medium. This was perhaps due to apical dominance of shoot tip and axial bud of nodal segments. NAA is indispensable for root production of in vitro culture of Stevia. Shoot length during root production was the highest on the medium where the highest number of roots was recorded perhaps due to higher amount of nutrient absorbed by higher number of roots.
