Effect of Some Fungicides on Helminthosporium Oryzae

The inviro activity of eight fungicides against Hellminthosporium oryzae was evaluated on their ability to inhibit spore germination and mycelial growth. Maximum inhibition of spore germination was obtained with Di.hanc M.45 and Dithane M-22, Dithane Z-78 and Dithane M-22 arrested the mycelial growth most effectively

Objectives:

To find out the effect of eight fungicides against Helminthosporium oryzae

Methodology:

The commonly practiced spore-gerrnintion and mycelial growth inhibition tests were used in the screening program. The chemicals used in these tests are collected from local firms and their details information are given below. Spore-germinatlon test :---In general the method of Abeygunawardena and Peiris (1958) was practised with slight modifications. Fungicidal preparations of four different concentrations 120,50,[00,200 ppm) of double strength of the test concentrations were prepared in distilled water. The spore suspension was prepared from 10 day old cultures and the spore concentration was adjusted to approxmately 10-15 per optical field (430). Equal volume of this preparation and the spores suspension were mixed In a groove slide and thus the strength of the fungicide was reduced to test concentration. A sample of 0.02 ml of this mixture was pipetted on a clean cover slip in the form of a drop. The cover slip with the fungal spore suspended in the solution was then put on a Van Tieghem cell in inverted position so that the drop remains inside the ring without touching its edge. The rings were fixed on the petridish (14'5 ern) by sealing with wax. Moist blotting paper was put in between rings and on the inner side of the lid of the petridish to maintain humidity. Each fungicide tested in four concentrations and three replica- .tions were used in each treatment. The cover slips containing the drops of diffe- rent concentrations of each fungicide along with the cover slip of distilled water (con- trol) were put in one petridish. After four hours the cover slip was collected and placed on a drop of lactophenol on a clean glass slide. Lactophenol was used to kill the organism. The slide was then examined.

Achievement/Findings:

Spore-germination test: Spore germination was inhibited by all fungicides at higher concentrations. Dithane M-45, Dirhane M-22 were statistically superior to other materials in inhibiting the spore germination. The effectiveness of Dithane M45 at a higher concentration on the growth of Helmintho- sporium oryzae was also confirmed by Kumar et al. (1973). Kasumiu and Blastin were very effective against Piricularia oryzae (Anonymous 1964 ; Ashrafuzzaman and Frederiksen 1970). But in this test they were less effective. These chemicals were obtained from Japan and were over one year old. This may be attrrbute d to this non effectiveness of these chemicals. Some inhibition of spore-germination was obtained with Benlate . A similar result was also obtained in case of Piricularia oryzae ( Ashrafuzzaman and Frederiksen (1970). Mycelial growth inhibition test: Mycelial growth was not inhibited at 25 ppm concentration of any of the fungicidal preparations. Dithane Z-78 most efficiently inhibited the mycelial growth. Duhane M-22, Dithane M-45 and Blasticidin- S had smaller yet measurable inhibit.on zones compared to that of other materials.
