The most conclusive test of sperm quality, the fertilization test of silver barb, Barbodes gonionotus, sperm was done. The males were stripped before the ovulation of female to make the fish breeding protocol easier. Modified Ringer's solution and modified Hank’s balanced salt solution were used as extenders to dilute milt. The diluted milt kept for one hour at room temperature (25°C) with 100 % viability and motility. The ratio of milt to extenders was 1 : 9. The significant effects of the extenders were observed in ability to fertilize eggs, no effect was observed on the embryonic development in the egg shells. The female silver barb was induced to breed in Linpe's method.

To find out the efficacy of the two most common  to retaining the ability of sperm for the fertilization of the eggs of the silver barb, B. gonionotus, for short time in normal temperature, which helps to make the fish breeding protocol easier.

The study was conducted by several steps, such as milt collection, measuring milt density, estimate sperm concentration, determining light absorption by milt in dilution 100 times, measuring sperm head size, observing sperm deformities, sperm viability and sperm motility. Finally the ability of sperm to fertilize eggs and embryonic survival up to larval movement were tested statistically. Three male brood fishes (22.5 ± 0.41 cm in length; 125.00 ± 8.17 g in weight) were selected on the basis of maturity and ease in the release of the milts. The broods were conditioned at circular cistern in a hatchery for more than 48 hours to evacuate the intestines. The milt was collected from the broods by pressing gently on their abdomen. Before milt collection, the broods were anesthetized by benzocaine (0.1 to 0.25%) by spraying directly on the gills. After collecting the milt in petridish, 0.1 ml of milt (Vs) was removed by a micropipette. The weights of petridishes were taken before and after removing the milt. The weights of the collecting milts (W s) were recorded by subtracting the two weights. Therefore, the density of milt was D (g/ml) =Ws/Vs. Number of sperm/ml = {(Nsp)/(Nsq )}* (D* 4*106).  The milt was diluted in counting solution in 100 times. The light absorption was recorded in UNICAM 8675 spectrometer at 420 μ weave-length. The per cent of fertilization (F%) was calculated by using the following formula (Deppert et al.1987): F%= {(E T)/(S T+ O)}* 100. Where, E T = Number of transparent eggs in the sample, and S T + O = Number of eggs in the sample both transparent and opaque. Again, the per cent of active larval movement (aLM%) at the time of larval movement was calculated by using the following formula: aLM%= {(EA )/(SA + Na)}* F%.  Where, EA = Number of eggs showing active larval in the eggshells of the sample. S A + Na = Number of eggs in the sample both having active and non-active larva in their shell. The data were analyzed by using a F-test. A t-test was also done at 0.05 confidence level. Analyses were done under statistical software programme IRRISTAT (version 4.0) on a computer. The programme was developed by the Biometrics unit of International Rice Research Institute (IRRI), Makati City, Metro Manila, the Philippines.

In this study, it was tried to simplify the fish breeding process. At first milt was collected from male without using any hormone. Then, the highly concentrated milt was diluted with the extenders, which helped sperms to mix with eggs properly. The extender helps not only diluting milt but also preserved the sperm for a short time. The preserving ability of the extender allowed striping male before the ovulation of female, which technically helped the breeding process. Generally, the ovulation and spawning of carp female occur at the same time and the female releases all eggs at a time in a very short time. So, the collection of the eggs is somewhat difficult in carps. In this system, full attention could be given to the egg collection from the female. Moreover, it would also save manpower. Again, the female was induced for breeding in Linpe’s method. The Linpe's method has been successfully used in silver barb spawning since 1988 in Thailand. Moreover, several tests were carried out which could be done without the female brood with full confidence for the collection of the milt in any season when female brood would not be ready. According to assessing the efficacy of extenders, fertilization experiments may become undependable (due to varying egg quality), expensive (brood fish maintenance) and impractical (annual breeders with short breeding season). However, further works are required to know the efficacy of the extenders retaining the ability of sperm to fertilize eggs and standardize the technique.
