Effect of Culture Filtrates of Four Common Seed Fungi on the Germination and Seedling Growth of Rice

Abstract/ Executive Summary:

Four common seed fungi namely Aspergillas niger, Aspergillus flavus, Fusarium sp. and Curvularia sp., isolated from stored rice seeds were used for determining the effects of their culture filtrates on rice seed germination and development of root and shoot length of the seedling. The cultivars used in this experiment were Hashikalmi, Morich-Bati, BR-3 and BP-3. The culture filtrates of A. favus and Fusarium sp. had inhibitory effect on the germination of seeds of all the selected cultivars, while the culture filtrate of A. niger affected the germination 0f Morich-Bati seed only. Decreased root and shoot length of the seedlings were recorded in Hashikalmi, BR-3 and Morich-Bati and only shoot length in Hashlkalrni and BR-3 when seeds were treated with the culture filtrates of A. Flavus and Fusarium spres pactively.

Objectives:

To the effect of culture filtrates of four common seed fungi on the germination and seedling growth of four selected cultivars of rice.

Methodology:

Four common seed fungi isolated from stored rice grains namely Aspergillus niger van Tiegh, Aspergillus Ilevus link, Fusarium sp, and Curvufaria sp. were in- cluded for the present study. The stock culture of the selected fungi in pure form were maintained in PDA medium. Pure culture of each of the fungi were prepared in sterilized Czapek's liquid medium. To prepare the pure culture, a loop full of spores from each of the selected culture was transfered to each 500ml Erlenmyer flask containing 250ml medium. After 10 days of incubation at room temperature, the culture media were filtered through Whatman filter paper No 42 to obtain the culture filtrate. The culture filtrate thus, obtained for each fungal isolate was tested for seed germination and seedling growth of four rice cultivars (Hashikalmi, Marich. Bati, BR-3 and BP-3). One hundred sterilized seed (treated with 1: 1000 mercuric chloride solution for two minutes and subsequently washing them with three changes of sterile water) from each selected rice cultivar were immersed in 20ml culture filtrates of each funqus separately. Two control sets, one with sterilized distilled water and other with blank liquid medium were also maintained. After 20 hours of treatment the seeds were removed from the culture filtrates and washed thorouqhlv with distilled water repeatedly. The seeds were placed in peteridishes, containing pre-autoclaved moist sand. The percentage of germination of seeds and the length of the root and shoot were recorded after 120 hours.

Achievement/Findings:

The findings of the present works is also more or less similar with the results obtained by different authors with other crops. Very little information is available regarding the effect of culture filtrates of seed fungi on the germination of rice seeds. To the knowledge of the authors no work has been done on this area tn the country. Therefore, further detailed investigation are necessary to draw any definite conclusion regarding the low germination of rice due to the associated seed fungi.
