A pot experiment was conducted at the BSMRAU, Gazipur under polyvinyl shed for exploring the variation of N use efficiency in mungbean genotypes GK3 and IPSA 25 under two N levels. Nitrogen was applied in nitrate form from Hoagland 1 (supplying 34ppm N) and Hoagland 3 (supplying 154ppm N) solution. Among the genotypes, GK3 had higher N concentration (%) in different plant parts over time till maturity under both high and low N levels. GK3 was relatively efficient at both the levels of N supply than IPSA25. IPSA 25 was relatively low in uptake efficiency, but highly efficient in utilizing N in grain production (Gw/Nt) at high N level. It was also above average in the fraction of total N that was translocated to grain (Ng/Nt). As such, IPSA 25 may be considered as highly potential genotype for low nutrient environments.

The experiment were to look for genotypic differences in the pattern of N absorption over the growth period

 (a) total N uptake (by monitoring N removal from solution and by examining total N accumulated by the plants), and

 (b) partitioning of total N in plant parts

Two mungbean genotypes viz. GK3 and IPSA 25 formed the treatment variables in the experiment. The genotypes were raised under two nitrogen levels (high N - 154ppm, low N - 34ppm). Hoagland 3 solution [3.5 mM Ca (NO3)2, 5 mM KNO3. 1 mM KH2PO4, 0.99 mM MgSO4, 19 M Fe-citrate and microelements] made of 154ppm N (high N level) and Hoagland 1 solution [0.5 mM Ca (NO3)2, 5 mM K2SO4, 0.5 mM Ca(H2PO4)2, 2 mM CaSO4. 0.99 mM MgSO4, 19 µM Fe2O3, and micronutrients] made of 34ppm N (low N level) were used as source for nitrogen. In all cases, nine uniform seeds of each genotype were germinated in net bag filled with vermiculite and inserted in the upper portion of 30 cm long polyvinyl chloride (PVC) pipe sections. The seeds were sown on 9 September 2001. When root extension was sufficient to allow satisfactory transfer (c. 14 day), 9 uniform plants were transferred to individual containers holding Hoagland 1 solution and Hoagland 3 solution which were reduced to 3 plants per 20 liter container at 21 DAE. Plants were supported in the upper portion by bamboo sticks. Two extra pots were monitored to ensure that no N was lost from solution by denitrification. The solution pH was maintained at 6.5 by adding required amount of 4N H2SO4. The solution was changed twice in a week. One of the seedlings was harvested at 35 DAE and another at 49 DAE and the remaining one was harvested at maturity (c.63 DAE). Plant height, root volume, leaf area, total root length was measured just after harvest. One plant at a time from each pot was harvested at 21,35,49, and at 63 DAE for growth analysis and determining N use efficiency. Harvested plants were separated into root, stem, leaf, petiole, and reproductive organs and were dried at 70°C till constant weight was attained. Dry weight of individual plant parts was recorded, ground in laboratory mill and passed through 0.18mm sieve for determining N content in each plant part at vegetative, flowering, pod filling and at maturity stage by micro-Kjeldahl method. N uptake was determined by multiplying the content by dry weight. The following data, all in mg/plant, serve for analysis: Gw = grain weight, Nt = total above ground plant N at maturity, Nv = total above ground plant N at flowering, Na = Nt- Nv = above ground plant N accumulated after flowering. and Ng = N accumulated in the grain at harvest. Analysis of variance was computed for these rive traits. Contribution of the various components to variation among genotypes in efficiency of N use at each level of N fertilizer was determined by sums of squares and sums of products of log ratios of genotype means as outlined above in the previous section.

Among the two genotype,IPSA 25 was relatively low in uptake efficiency, but  was highly efficient in utilizing N  in grain production (Gw/Nt) at high N level.
