Screening of Organic Composts for Mass Culturing of Trichoderma Harzianum to Be Used Against Soil-borne Pathogen Sclerotium Rolfsii of Lentil

Abstract/ Executive Summary:

The experiment was conducted to observe the suitability of organic compost and vermi-compost for mass culturing of bio-control agent, Trichoderma harzianum and its effectiveness against foot and root rot disease of lentil caused by Sclerotium rolsfii and Fusarium oxysporum. The formulated T. harzianum cultured in two different compost viz. organic compost and vermin-compost is designated as Tricho-organic-compost and Tricho-vermi-compost. The present study revealed that soil amendment with Tricho-vermi-compost and Tricho-organic-compost are considered to be the best treatments in reducing seedling mortality and in increasing yield of lentil. Seed treatment with chemical fungicide such as Provax showed better performance against the disease and its effect at per the soil amendments with organic compost and vermin-compost in reducing seedling mortality and increasing plant growth and yield of lentil.

Objectives:

The present research was under taken to find out the effective formulation of T. harzianum against foot and root rot disease of lentil.

Methodology:

The experiment was conducted in the field of Plant Pathology Division, Bangladesh Agricultural Research Institute, Joydebpur, Gazipur. There were 6 treatments viz. (i) Seed treatment with Provax (ii) Soil amendment with organic compost (iii) Soil amendment with Tricho-organic compost (iv) Soil amendment with vermi-compost and (v) Soil amendment with Tricho-vermi-compost and (vi) Untreated control. The unit plot size was 2.5 m x 3m. RCB design was followed with 3 replications. T. harzianum was multiplied in the mixture of Grasspea and wheat bran with mustard oilcake substrates. The formulated Trichoderma was used to mass multiplication in two different compost viz. organic compost and vermin-compost and its designated Tricho-organic-compost and Tricho-vermi-compost. T. harzianum were cultured in PDA media and spores were harvested 10 days old cultured and were used for seed treatments. The soil was inoculated with S. rolfsii colonized substrates @ 100g/m² soil before seed sowing. Tricho-composts were added @ 3 t/ha after inoculation with pathogens. Seeds were treated with Provax @ 2.5 g/kg seeds before seed sowing. BARI Masur-5 variety was used in this study. Data were collected on seedling mortality; shoot height, shoot weight, root length and root weight. The yield of lentil was also recorded. Data were analyzed by MSTATC program. Means were computed by Duncun’s New Multiple Range Test (DMRT).

Achievement/Findings:

Significant difference was observed among the treatments. Soil amendment with Tricho-organic-compost and Tricho-vermi-compost gave the higher shoot height; shoot weight, root length and root weight followed by seed treatment Provax and soil amendment with organic-compost and vermi-compost compared to control. All the treatment gave significantly higher seedling emergence than control. The highest seedling mortality 46.67% was found in control treatment. Soil amendment with Tricho-vermi-compost gave the lower seedling mortality 10.67% followed by soil amendment with Tricho-organic-compost and seed treatments with Provax, where seedling mortality was 11.67% and 13.33%, respectively Soil amendment with organic-compost and vermi-compost also observed significantly lower seedling mortality with 16.67% and 20.67% compared to control but significant differences among the treatments was not observed. Soil amendment with Tricho-vermi-compost reduced 76.29% seedling mortality compared to control followed by soil amendment with Tricho-organic-compost and seed treatment with Provax where the reduction seedling mortality was 74.07% and 70.38%, respectively than control. Soil amendment with organic-compost and vermi-compost reduced 62.96% and 54.07% seedling mortality compared to control. Soil amendment with Tricho-vermi-compost and Tricho-organic-compost gave higher yield of lentil at 1943 and 1896 kg ha^-1, respectively followed by soil amendment with vermi-compost, seed treatment with Provax, and organic-compost where the yield was 1505, 1500 and 1484 kgha^-1, respectively. The lowest yield of lentil 1078 kgha^-1was recorded from the control treatment. Yield was higher 44.52% compared to control due to application of Tricho-vermi-compost -2 where as it was 43.14% due to application of Tricho-organic-compost. Soil amendment with only vermin-compost gave higher yield 28.37% where it was 28.13% and 27.36% by seed treatments with Provax and soil amendment with organic-compost, respectively compared to control. From this study it is observed that two compost viz. vermin-compost and organic compost are the suitable media for mass culturing of bio-control agent T. harzianum and soil amendment with cultured Trichoderma designated as Tricho-vermi-compost and Tricho-organic-compost is to be the best treatments for reducing seedling mortality and increasing plant growth and yield of lentil. Seed treatment with Provax also better treatment for management of foot & root rot disease of lentil. Soil amendment with organic and vermin-compost alone also have significant effect on foot and root disease of lentil compared to control. In the present study inhibition of seedling disease caused by soil borne fungal pathogens by Trichoderma harzianum.

From this it may be concluded that soil amendment with Tricho-organic-compost and Tricho-vermi-compost are to be the best treatment for management foot and root rot disease of lentil and getting higher yield of lentil than control. Seed treatment with chemical fungicide Provax also better treatment for management foot and root rot disease of lentil.
