Effect of Seed Coat and Kernel Extracts of Different Plants on the Lesser Mealworm, Alphitobius Diaperinus (panzer) (coleoptera: Tenebrionidae)

Abstract/ Executive Summary:

The chloroform and methanolic extracts of seed coat and seed kernel of some plants, e.g. Caesalpinia bonduc (natai), Mucuna pruriens (alkushi), Adenanthera pavonina (rakta chandan), Terminalia bellirica (bahara), Syzygium cumini (kalojam) and Myristica fragrans (jayfal) were tasted for mortality against the adults of the lesser mealworm, Alphitobius diaperinus(Panzer). Majority of the extracts showed significant mortality effects on the beetle. Seed kernel extracts showed a higher degree of toxicity on the test insect, as compared to the seed coat extracts. Methanolic extracts of all seed parts showed higher toxicity on the test insects

Objectives:

In this investigation, insecticidal activity tests of six indigenous plants were carried out on the lesser mealworm to evaluate the efficacy of the seed coat and seed kernel extracts of the test plants as a possible source of potential secondary metabolites to be used as environment friendly pest control agents.

Methodology:

The fresh seeds of C. bonduc (natai), M. pruriens (alkushi), A. pavonina (rakta chandan), T. bellirica (bahara), S. cumini (kalojam) and M. fragrans (jayfal) were collected from the Botanical Garden, Rajshahi University and the identification of voucher specimens were confirmed at the Taxonomic Section, Department of Botany, University of Rajshahi, Bangladesh. The plant parts viz., seed kernel and seed coat were chopped into small pieces, dried under internal shade and powdered using a hand grinder separately. The seed kernel and seed coat powder were extracted with chloroform and methanol (BDH, poole England) using Soxhlet's apparatus according to Feuerhake and Schmutterer (1982). The extracts thus obtained were stored in a refrigerator at – 20⁰C with proper labeling. The chloroform and methanolic extracts of the experimental plants were tested for mortality against the adults of A. diaperinus through dose- mortality with various doses on the surface of Petri dishes (6 cm. diam.) where the test insects were released to observe for mortality or any kind of abnormality due to the effect of the extracts compared to the controls. Data were subjected to statistical analyses according to Finney (1947) and Busvine (1971), and the dose-mortality relationship was expressed as the median lethal dose (LD50)

Achievement/Findings:

The present results clearly demonstrate that both the chloroform and methanol extractives from the seeds of different plants have significant insecticidal activities. The contact toxicity of the extract on the test insect in terms of LD50 (median lethal dose) of the extracts in two solvents from seed kernel and seed coat were not similar. In general, all seed kernel extracts showed higher degree of toxicity on the test insect, as compared to the seed coat extracts. It is evident from the present investigation that the seed extracts of different plants are significantly active against A. diaperinus. The seed extract produced the highest mortality at lower concentrations. More comprehensive works are very much to be solicited for their effective use, especially in medicine and agriculture.
