An experiment was carried out at Hajee Mohammad Danesh Science and Technology University, Dinajpur, Bangladesh during March to May 2016. Three types of containers and three levels moisture content i.e. 9.75, 12.25 and 14.30% were used as the treatment. Mustard seeds with 9.75% initial moisture level were found in good condition in respect of germination, less number of abnormal and dead seedlings, better shoot and root length, vigour index and poor fungal incidence. The highest germination was observed 80.14−87.74%, while the lowest percent of abnormal seedling (8.64−12.04%) and dead seedling (3.33−8.27%) was found in the seeds stored with 9.75% initial moisture content at different storage periods. Fungal incidence was also found less in the seeds stored with 9.75% initial moisture content. Among three storage conditions, plastic container was showed the advantage for mustard seed storage compared to polythene bag and gunny bag. The seed of gunny bag was found inferior in quality for all parameters at all storage period. So, the results revealed that optimum moisture content and ideal storage condition has the greatest benefit on quality seeds. It is concluded that plastic container with 9.75% initial moisture content is the best for storing the mustard seed with higher quality.

To know the effect of initial moisture content and different storage container on the quality of mustard seed.

The experiment was conducted at Hajee Mohammad Danesh Science and Technology University, Dinajpur, Bangladesh during March to May 2016 to study the effect of initial moisture content and storage container on the quality of mustard seed. The experiment was laid out in Completely Randomized Design with three replications. Three seed containers (plastic container, polythene bag and gunny bag) and three seed moisture levels (9.75, 12.25 and 14.30%) were included in the study. Mustard seeds (14.65% moisture level) collected from the market were equilibrated to moisture levels (14.30, 12.25 and 9.75%) by sun drying and stored for two month in plastic container, polythene bag and gunny bag at room temperature. During the storage period, hundred seeds samples were taken at every 15 days interval from the each container for determination of the germination percentage, abnormal and dead seedling, shoot and root length, vigour index and fungal incidence. Moisture content was determined by using high constant temperature oven method following International Rules for Seed Testing (Anonymous, 2000). From each container three gram seeds were taken. After grinding the seeds in grinding mill, the weighed ground materials were poured separately in a small container with cover and kept in an oven maintained at a temperature of 125-1300C for a period of 2 hour. The moisture content of seeds (wet basis) was determined by the following formula (Anonymous, 2000). Germination test was conducted using sand as substratum. The sand was sieved to discard particles bigger than 0.8 mm and smaller than 0.05 mm in diameter. Rectangular plastic boxes were used to put the sand for every test. Seed was placed on a uniform layer on moist sand and then covered to a depth of 10 mm with sand, which was left loose. 100 seeds were planted in each plastic tray and replicated 3 times. The plastic trays with seed were incubated at room temperature and irrigated at every odd day. After 8 days of sowing, germination percentage was recorded. The normal seedlings, and abnormal seedlings and dead seedlings were classified according to the prescribed rules given by ISTA (1996).

For vigour index data was recorded on the germination up to 10 days of sowing. Then root and shoot length from the seedling were measured for calculating vigour index. Vigour index (VI) was calculated by the formula reported by Baki and Anderson (1973) showed as Vigour index (VI)=Germination (%)×(Mean shoot length+mean root length) Seed borne pathogens associated with the seeds in samples was detected by blotter method by following international rules for seed testing (ISTA, 1996). Most of the associated pathogens were detected by observing their morphological traits on the incubated seeds on blotter paper. For proper identification of fungi, temporary slides were prepared from the fungal colony and observed with compound microscope and identified following the key outlined by Booth (1971) and Singh (1982). The collected data were analyzed by analysis of variance using a MSTAT-C statistical computer programme and the mean values were separated by Duncun’s Multiple Range test (DMRT).

Mustard seeds could be stored in plastic container and moisture level at 9.75 % showed comparatively better performance than other containers (poly bag and gunny bag) and moisture content (12.25% and 14.30%) in case of germination, less number of abnormal and dead seedling, shoot and root length, vigour index and poor fungal incidence at the storage condition.
