Plant Pathology Division, BARI, Gazipur

An attempt was taken for the management of bacterial wilt of brinjal caused by Ralstonia solanacearum by using some bectericides at Bangladesh Agricultural Research Institute (BARI), Gazipur-1706.  In the in-vitro trial we observed that Bleaching powder, Calcium Nitrate and Bismerthiazol (Bactrol) did not show any effect against R. solanacearum, when Streptomycin sulfate+ Tetracycline hydrochloride, Streptomycin sulfate, Bismerthiazole and Oxytetracycline hydrocloride completed blocked bacterial growth. Based on the preliminary laboratory evaluation, Streptomycin sulfate+ Tetracycline hydrochloride, Streptomycin sulfate, Oxytetracycline hydrochloride, Tetracycline, Bismerthiazole were selected for the control of bacterial wilt disease in the field experiment. In the field trial, among the different treatments Bismerthiazole, Tetracycline and Streptomycin sulfate+ Tetracycline hydrochloride was appeared to be superior in controlling the wilt of brinjal and also significantly increased the yield

The experiment was undertaken to find out the effective bactericides in controlling bacterial wilt of brinjal

The study was conducted at the Plant Pathology Laboratory, Bangladesh Agricultural Research Institute during Rabi season, 2018-19. The field experiment was carried out in the field of Plant Pathology Division, BARI, Gazipur, Bangladesh. Bacterial culture and inoculum preparation: R solanacearum was collected from the severely infected from Jessore. The bacterium was sub-cultured on nutrient agar YPSA medium at 28°C for 48 h. The bacterial cells were flooded with sterilized distilled water (SDW) and scrapped off the NA plate with a sterilized cotton swab to make a bacterial suspension of (OD600= 0.3) in a spectrophotometer (Wai et al., 2013 and Lin et al., 2014). This suspension was used for in-vitro bacterial inhibition tests as well as for inoculation of healthy plants during transplanting experiments. Six bactericides were tested in the lab for bioassay and artificially infected seedlings will be tested for the field trial. Three sprays will be done from the onset of disease at 15 days interval. In-vitro assessment of some chemicals against R solanacearum: six chemicals viz Bleaching powder (100 g/L), Calcium Nitrate, Bismerthiazole, Streptomycin sulfate+ Tetracycline hydrochloride (300 ppm), Streptomycin sulfate (0.2%) and Oxytetracyclinehydrocloride (300 ppm) were tested for their ability to inhibit the in-vitro growth of R. solanacearum using well diffusion method (Yenjerappa, 2009). A bacterial lawn was prepared by pouring 100 µL of the bacterial suspension (1.5×108 cfu/mL) on NA plates and spreading uniformly (Balestra et al., 2009 and Hindi et al., 2013). Each well was loaded with 20 µL of each chemical solution and control was loaded with 20 µL distilled water. The plates were incubated overnight at 30°C and the zones of inhibition were measured at 24 hours, 48 hours and 72 hours across the well with a transparent plastic ruler. The field experiment was conducted with five chemicals using a Randomized completely block design (RCBD) with three replications. Calculation of disease incidence: To calculate the percent disease incidence, the number of plants wilted per total number of plants was counted. Percent disease incidence was calculated by the following formula as described by Mansoor et al. (2007):

                                                           Number of plants wilted

                       % Disease incidence= --------------------------------- X 100

                                                          Number of plants observed

Data analysis:  The experiment was done using Completely Randomized Design (CRD) with eight replications. Data collected during the experimental period were tabulated and analyzed using Statistical software R and treatment means were compared with the least significance difference (LSD) test at p=0.05. Six chemicals were tested in the field trial viz.
