To develop an integrated management package for soft rot of potato, in February 2019, BARI Alu-72 were collected from Breeder seed production center, Debiganj, Panchagarh district. The tubers were artificially inoculated with previously characterized Pectobacterium carotovorum subsp. carotovorum PKI2 isolate and ten different treatments including chemical, biochemical and biological control agent were used to reduce the disease severity (%) of the soft rot infected potato tubers. Among them, treatments containing 3% chitosan (T4) and Trichoderma sp. (T5) reduced area under disease progress curve (AUDPC) significantly for soft rot severity in comparison to the non-treated control.

To develop an integrated management package for soft rot of potato

At the end of February, 2019 BARI-alu 72 was collected from the Breeder seed production center, Debiganj, Panchagarh. Previously characterized Pectobacterium carotovorum subsp. carotovorum PKI2 isolate (GenBank accession-KX098357) was used to inoculate the tubers. This isolate was streaked on luria-bertani agar (LBA) medium and incubated at 28°C for 3 days. Bacterial concentration was confirmed by dilution plating (108CFU/mL) and adjusted to an optical density of 0.2 (OD600). Seven randomly selected tubers of BARI-alu 72were washed with 0.5% sodium hypochlorite and dried for 1 hour. The tubers were cut in half and data were taken for 2 weeks. Preparation of Trichoderma harzianum inoculum: Bacterial suspension (107conidia/mL) was prepared by adding sterilized distilled water to 7-days-old T. harzianum BTH-N1 cultures grown on PDA, incubated at 28° C for 7 days. Preparation of chitosan solution: Water-soluble chitosan was sourced online from China (Qingdao Yuda Century Economy and Trade Co. Ltd, China). Sterilized distilled water was used to make 1% and 3% chitosan solutions (1g/100mL, 3g/100mL), which were kept for 12 hours at room temperature to form a uniform solution (pH 5.5). Full size sprouted potato tubers were surface sterilized (using 70% ethanol for two min) and were dried under laminar airflow for 10 min. Potato tubers were cut into half and submerged in the chitosan solution for 30 min, then air-dried.

This experiment was conducted in the lab of the Plant Pathology Division, BARI in Plastic boxes.  And the temperature ranged from 25-28°C. The treatments were as follows: T1= 50 ppm chlorinated water, T2= 100 ppm chlorinated water, T3= 1% chitosan solution, T4= 3% chitosan solution, T5= Trichoderma sp., T6= 50 ppm chlorinated water+3% chitosan solution, T7= 100 ppm chlorinated water+Trichoderma sp., T8= 1% chitosan solution+Trichoderma sp., T9= 1% chitosan solution+ Trichoderma sp. + 50 ppm chlorinated water and T10=Control were evaluated in a complete randomized block design with three replications. Seven randomly-selected tubers were mock-inoculated with sterilized distilled water served as a negative control. Data were taken after 7 days of inoculation to 14 days at a 7-day interval. Disease severity was measured on a 0-3 scale; 0= no symptoms of soft rot infection, 1= less than 50% of the tubers have soft rot symptoms, 2= more than 50% tubers have soft rot symptoms, 3= tubers are completely rotten. The area under the disease progress curves (AUDPC) was calculated based on soft rot percent severity according to the formula, where is the rating at each evaluation time and (ti-ti-1) is the number of days between evaluations.

Data analysis: R statistical software was used for the analysis of variance (ANOVA). Differences in means were assessed using the Least significant difference (LSD) with an error rate of α=0.05.

Treatments containing 3% chitosan (T4) and Trichoderma sp. (T5) reduced AUDPC significantly for soft rot severity in comparison to control. All the other treatments except T7 (100 ppm chlorinated water+Trichoderma sp.) reduced AUDPC compared to control though they are not significantly different than the control treatment.
