An experiment was carried out at the Pomology Division, Horticulture Research Center, BARI, Gazipur during the flowering season of 2018-19 owing to study the pollen viability of litchi during storage. Male flowers of BARI Litchu-2 were collected at anther dehiscence stage; time between 8 and 10 a.m. Collected pollens were stored at 4 different storage conditions such as; at room temperature (25-300C) in petridish, at room temperature in desiccator, refrigerator (5-70C) and deep freeze (-20±20C). Fresh pollen showed 43.37% viability, which had 26.68% viability after 7 days of storage in petridish. After 7 days of storage, 31.43% viable pollen was noticed when pollen was stored in desiccator at ambient temperature. Pollen viability under refrigerated condition lasted up to 60 days and it was observed 23.18%. At deep freeze (-20±20C) condition, pollen remained viable up to 60 days, which was 32.02%. Pollen viability was tested through in-vitro germination for all the storage conditions. Pollen stored in Deep freeze remained viable up to 60 days, which exhibited 29.84% germination at the same condition.

To select the litchi genotypes with the objectives of enhancing the quality of collected pollen, maximizing survival on transport, storage and thawing, optimizing retesting of pollen viability in the lab, and safe transport to enable field pollinations.

An experiment was conducted at the Pomology Division, Horticulture Research Center, BARI, Gazipur during the flowering season of 2018 and 2019 to study the viability of litchi during the storage. The present study elaborates an improved method for pollen collection, transportation over a long distance before storage and transportation back to the site of the female parent in a viable condition. Male flowers of BARI Litchu-2 were taken for this study. Data were analyzed by using Excel program.

Pollen collection and storage

Male flowers of BARI Litchu-2 were collected at anther dehiscence stage, time between 8 and 10 a. m. After sufficient number of flower collected, anthers separated from the flower and dried into a desiccator with silica gell for 2-3 days. Then collected pollens were sieved by 80 mesh sieve and then pollen grain transferred to a cryovial for storage. These vials can be transported in an ice box (holding ice at about 50C) within 2 days of collection. Collected pollens were stored at 4 different storage conditions such as at room temperature (25-300C) in petridish, at room temperature in desiccator, refrigerator (5-70C) and deep freeze (-20±20C).

Pollen viability test

Litchi pollen viability was tested by using acitocarmine staining test and in-vitro germination test. A single drop of acitocarmine solution was taken on a slide and pollen grain placed on it with a needle and then covered with a cover glass and observed under the microscope.In vitro germination of pollen grains was observed by placing in a sugar-ager media. In abeaker containing water, 20% sucrose and 1% ager was taken and heated until the ager dissolves completely. Boric acid @ 0.1% was also added to it. When the ager dissolved completely then media was poured into 50 ml petridish and kept in a dark place in room temperature. When the medium became cool, then pollen grains were placed with a needle on the medium and kept again in room temperature. Then the slides were placed under a microscope. Germinated pollen was counted from three to four different fields from a single slide and percent value was calculated.

Pollen viability and in-vitro germination were investigated in this study. Maximum pollen viability was recorded from the fresh sample. Duration of pollen viability was obtained up to 60 days when pollen kept in refrigerator. Pollen viability was tested by in-vitro germination for all the storage conditions and pollens stored in deep freeze condition exhibited higher percentage of germination, which remained viable up to 90 days. In this study, second year observation is going on and more trial is required for final recommendation.
