Abstract/ Executive Summary:

A total of 210 broilers birds and 280 eggs were collected from different markets of poultry rich area of seven divisions in Bangladesh in order to detect sulfonamide and ciprofloxacin residue in these items. For bacteriological examination, to fund only as the probable causes agents when drugs were used, a total of 60 samples of litter, feed, water and liver swab were obtained from different markets of Sirajgonj districts. Beside these, 24 broilers were also collected perform post mortem for various organ at to carry out serological examination as well as detection of antibiotic residues. Higher positive cases were detected for ciprofloxacin than sulfonamide, while highest mean of both antibiotics residue level was found in egg. Compare to the mean residues in kidney was and in case of tissue samples mean residue in kidney are higher than the other organs followed by liver, breast and thigh muscle respectively for both antibiotics. On the other hand the birds show negligible serological response with the commercial antigens but the water, feed and the litter were positive for E coli and salmonella spp indicating that antibiotics used while keeping the study for longer period. Such a practice may be a source of residual antibiotic in finished broiler.

Objectives:

2. To fund out the possible cause of drug residue.

Methodology:

Collection of Sample: Detection of residue in egg and broiler meat- A total of 210 broilers and 280 eggs were collected from different markets of poultry rich area in seven divisions of Bangladesh. Identifying the possible causes of drug residue while 24 broilers collected from different markets of Serajgonj district. Detection of antimicrobial drug residue with ELISA. Sample preparation: Solution-1: 1 x Sample extraction Buffer, 1 volume of 10 x Sample Extraction Buffer was mixed with 9 volumes of distilled water. Solution-2: 2 M NaCI Solution, 11.69 g sodium chloroxid were diluted with deionized water to 100 ml to prepared 2M sodium chloroxid solution. Tissues sample preparation for ciprofloxacin residue detection, Tissues sample preparation for sulfonamide residue detection, Preparation of egg sample for detection of for ciprofloxacin and sulfonamide residue and Procedures for ELISA test.

Achievement/Findings:

It was showed that higher positive case was detected for ciprofloxacin than that of sulfonamide; highest mean of ciprofloxacin and sulfonamide residue level was found in egg and in case of tissue samples mean residue in kidney were higher than the other organs followed by liver, breast and thigh muscle respectively for both antibiotics. On the other hand, the birds showed negligible serological response with the commercial antigens but the water, feed and the litter were positive for the bacteria. The postmortem lesions (duodenal hemorrhage cloudy air-sac, pulmonary congestion and consolidation) suggested that the birds were infected with the bacteria and became sick. These broiler stockiest used antibiotics to keep the birds alive and to escape the infections. This may be one of the major sources of antibiotic in our finished broilers.
