Microsatellite Markers for Determining Genetic Identities and Relationship Among Maize Hybrids and Their Parents

Abstract/ Executive Summary:

DNA fingerprint was generated using six SSR markers on 22 maize genotypes including nine hybrids and 13 parental lines. A total of 35 alleles were identified using six primer pairs among the 22 genotypes with an average of 5.83 alleles per locus. Locus UMC1353 was the most informative among the six, which distinguished ten genotypes. A high level of genetic polymorphism allowed unique genotyping of 68.18% of the studied genotypes for unambiguous identification of 15 maize genotypes. amplification of a 209/198 bp, 222/220/214 bp, 214/209 bp, 228/222/209/201 bp, 214/147 bp, 228/217 bp, 220/217 bp, 217/214/198 bp, 217/209 bp and 222/214 bp band with UMC1353 primer pair could distinguish BARI Hybrid maize-2, BARI Hybrid maize-8, BARI Hybrid maize-9, BARI Hybrid maize-10, BARI Hybrid maize-11, BARI maize-7, BIL-20, BIL-31, BIL-79 and BIL-110 respectively while the absence of this band and presence of a 141/139/130 bp and 139/130/124/112 bp band with bng1197, could identify BARI Hybrid maize-5 and BARI Hybrid maize-6 respectively and so on. The experiment provides proof high levels of genetic polymorphisms among the maize genotypes. This findings reinforces the utility of SSR primers for providing unique genetic identities or fingerprints of various maize cultivars

Objectives:

To Identifyf SSR markers useful for distinguishing 22 maize genotypes,
To develope genetic fingerprints for these genotypes
To estimate of relatedness among the hybrids and their parents.

Methodology:

Seeds of the nine maize hybrid varieties and 13 parent materials were collected from the Plant Breeding Division of the Bangladesh Agricultural Research Institute. The seedlings were grown in small plastic pots. Bulked DNA was isolated from 2-5 fresh leaves of 15 days old seedlings using following the protocol DNA was confirmed using 1% agarose gel and quantified at 260nm spectrophotometrically (Spectronic® GENESYS™ 10 Bio). Six primer pairs such as bng1197, bng1257, UMC1160, UMC1177, UMC1292 and UMC1353 was developed . Amplification was carried out in an oil free thermal cycler (Techne, TC 312 thermal cycler). PCR products were confirmed by electrophoresis on 2% agarose gel.

Achievement/Findings:

The genetic difference and unique SSR band profiles that we could identify at variety level in the new adaptive ecology should be conserved and maintained to protect the maize genotypes from further manipulation and registration. The results of the present study could be applied as baseline information to maintain the appropriate identity of maize cultivars, and in broad sense to protect the plant varieties of Bangladesh.
