Effect of Different Sources of Arbuscular Mycorrhiza on the Performance of Brinjal Seedlings

Abstract/ Executive Summary:

A pot study on the effect of different sources of arbuscular mycorrhizal (AM) fungi on chilli seedlings was conducted in the net house of Soil Science Division, BARI, Gazipur during the fourth week of April to first week of June, 2003. One kg of silted soils were mixed with cow dung at 5:1 ratio was used pot-1 as the potting media. Soil based AM inoculum was used at the rate of 50 g pot^-1. Growth, biomass yield and nutrient uptake of inoculated seedlings were significantly higher than those of uninoculated seedlings. Such higher growth and nutrient uptake appeared to be due to beneficial effects of AM fungi. All the local AM inoculum sources were found almost equivalent and better than the exotic source in respect of biomass production and nutrient uptake. Inoculation with AM fungi helped to produce healthy and vigorous seedlings with faster growth. This would help to reduce nursery life and thereby cost of seedling production. The AM inoculated seedlings might also perform better in the field since the fungi could be carried over to the field through the colonized roots.

Objectives:

Methodology:

A pot study on the effect of different sources of AM fungi on brinjal seedlings was conducted in the net house of Soil Science Division, BARI, Gazipur during fourth week of April to first week of June, 2003. The silted soils were mixed with cow dung at 5:1 ratio was used as the potting media. The mixture was sterilized with 10 percent formaldehyde solution to inactivate the native AM fungi. One kg of this sterilized mixture was used per pot as the potting media. The experiment was laid out in RCBD with four replications. Effect of AM fungi collected from Jamalpur, Joydebpur, Ishurdi and Hathazari, their mixture and an Exotic source (from India) were studied along a Control on brinjal variety Kajla. Perforated black polythene bag was used as the pot. A hole of 6cm in diameter and 5cm in depth was dug at the center of the potting media. The soil based AM inoculum at the rate of 50g pot^-1 containing 105 to 120 spores 100g^-1 soil. A soil layer of about 1cm was spread on the inoculum layer. Brinjal seeds were sown in the soil layer above the inoculum to ensure penetration of the roots through the inoculum layer immediately after germination. The biomass yield and yield components of seedlings were recorded. The seedlings were harvested carefully by uprooting at the age of 43 days after sowing (DAS). Root samples were excised for AM colonization studies. The dried plant samples were ground and used for chemical analysis. Soil samples from the pot were collected during harvesting the seedlings for counting AM spore population. Hundred grams soil sample per pot was used to count the spore numbers. The spore numbers were determined by wet sieving and decanting method. To assess AM root colonization, the roots were processed.

Achievement/Findings:

Performance of seedlings Performance of brinjal seedlings with different sources of AM inoculum revealed that dry matter yield and yield components of the AM inoculated brinjal seedlings were found significantly higher than those of uninoculated seedlings irrespective of inoculum sources. There was much difference in dry matter yield of the inoculated and uninoculated seedlings. The local AM inoculum sources appeared to be superior to the exotic source. Among the local sources AM inoculum from Joydebpur produced the highest dry matter yield and yield components of the brinjal seedlings but they were identical to those with the other local sources. Mycorrhizal Development Effect of different sources of AM inoculum on root colonization by AM fungi and spore numbers in rhizosphere soils of brinjal seedlings revealed that as of the dry matter yield the highest AM root colonization was observed with Joydebpur source but it was identical to those with the other local sources and with their mixture. Among the inoculated seedlings the exotic source produced the lowest AM colonization. There was also some root colonization in the control plants with native AM fungi. This might be due to survival of some native AM fungi even after sterilizing the soil. The spore numbers in the rhizosphere soils was found identical among all the inoculated seedlings irrespective of inoculum sources. Rhizosphere soils from control plants also contained some AM spores because of some root colonization with the native AM fungi. A significant positive linear relationship between the AM root colonization and dry matter yield of brinjal seedlings was observed. Nutrient uptake Uptake of all the nutrients in the inoculated seedlings was found significantly higher than that in the uninoculated seedlings irrespective of inoculum sources and the difference was much higher between these treatments.
