Productin of Cellulolytic Enzymes and Their Degradative Effect on Jute Products.

Abstract/ Executive Summary:

A number of mesophilic cellulolytic fungi were isolated and maintained on PDA slants for enzyme production on wheat bran medium.

Objectives:

Utilization of biologically decomposed jute materials and wastes as boi-fertilizer.

Methodology:

Collection of samples : Normally rotten wood, degraded jute bags and bamboo were collected from in and around the Dhaka city. The samples were kept in new white poythene bags and were labeled properly. Isolation of cellulolytic microbes ( fungi) : Small pieces of each collected samples were soaked in sterile water and shaked well. A little from this extraced liquor was inoculated in solid agar medium distributed in petridishes. After an optimum incubation the isolated colonies of fungi were randomly picked up, purified through gradual transfer from one plate to another. The pure fungal strain was maintained on PDA slants in the refrigerator and sub – cultured at one month intervals. Screening of Microorganisms : For detecting the enzyme activities of isolated fungal strains, the fungi were inoculated in solid agar medium distributed in petriddishes containing CMC as sole carbon source. The medium contained all necessary ingredients to afford sufficienyt initial growth of the inoculum.

Ten fungal colonies were isolated.The isolates were primarily marked as cellulolytic one on the basis of their ability to grow on CMC medium.
