In Vitro Screening of Potato Cultivars for Relative Salt Tolerance

Abstract/ Executive Summary:

In vitro screening of five high yielding (Provento, Patrones, Meridian, Diamant, Dheera) and three local (Zaubilati, Lalpakri, Hagrai) cultivars of potato for salt tolerance at different concentrations of NaCl were conducted. To select the salt tolerant cultivar, physiological parameters including shoot length, leaf number, shoot fresh weight, shoot dry wet and number of rooted shoots were measured. Considering the characters under evaluation cultivars Patrones, Meridian and Zaubilati showed relatively better performance under salt stress.

Objectives:

1. To evaluate the salinity tolerance of potato varieties 2. To find out the suitable variety/varieties for growing in salt affected areas

Methodology:

Seeds of high yielding and local potato cultivars were collected from the Tuber Crop Research Centre and Debiganj of BARI. Healthy sprouts of those varieties were collected and surface sterilized with detergent for 10 min. and washed with running tap water for 15 min. Explants were then rinsed with 1 to 2 drops of Tween 20 with ethanol for 20 sec. and washed two times with distilled water in a laminar air flow cabinet. After that the explants were rinsed with 0.1% HgCl₂ for 30 sec. and washed three to four times with distilled water. Then the sprouts were cultured in potato micropropagation medium. Cultures were maintained and sub cultured under 16/8 hours photoperiod at 25 ± 1⁰C and propagated by sub culture every 4 weeks. Single nodes were then transferred to potato micropropagation medium containing different concentrations of NaCl (0 mM, 40 mM, 80 mM, 120 mM, 160 mM) with three replications in each treatment in a Completely Randomized Design (CRD). After five weeks, the plantlets were harvested and the physiological parameters including shoot length, leaf number, shoot fresh weight, shoot dry wet and number of rooted shoots were measured. The shoot fresh and dry weights were measured at the time of harvest and after 60 h at 60 ± 2⁰ C in oven, respectively. Results were statistically analyzed using ANOVA in MSTATC program and range test was conducted by LSD for comparison of means.

Achievement/Findings:

Five weeks after salt treatment of plants, physiological characters including shoot length, leaf number, shoot fresh weight, shoot dry wet and number of rooted shoots were measured. Significant variation was observed among the cultivars in different parameters studied. The maximum shoot length was observed in Zaubilati (4.57 cm) followed by Patrones (4.40 cm) and Meridian (3.45 cm). The highest leaf number (8.29) was recorded from Patrones followed by Zaubilati (6.80) and Meridian (6.40). Patrones also produced the highest shoot fresh weight (32.65 mg) followed by Meridian (19.25 mg). On the other hand, the highest shoot dry weight also produced from Patrones (1.90 mg) followed by Meridian (1.33 mg) and Zaubilati (1.21 mg). In the present study, it was observed that genotype responded differently in salt stress condition. Some variety produced roots at higher level of salinity. Lalpakri produced roots in highest salinity (160 mM) level. Provento, Zaubilati, Diamant and Hagrai produced roots up to 120 mM salinity level.
