Improvement of Jute Fibre by Using Microbial Enzymes.

Abstract/ Executive Summary:

Highest yield of extracellular xylanase ( 251.23 IU/ml) was obtained from the isolate BJBC-984(2) at 50% moisture content under solid state fermentation condition. It was observed that the effect of maximum pH value for the activity of crude xylanase was 5.5.Xylanase was found to be most active at 400C . The level of xylanase activity at 400C was approximately 250.97 IU/ml . Above 500 the activity of the enzyme decreased sharply. The enzymre obtained from the isolate BJBC – 984(2) retained 100% of its activity after 28 days of incubation at room temperature .The specific activity of xylanase of BJBC - 988 is barely over 200 and the specific activity of xylanase of organism BJBC-984(2), which is close to 400, is found to be better.

Objectives:

To modify the physical and chemical characteristics of jute abd allied fibres to improve their textile properties.

Methodology:

Microorganism BJBC – 984(2) and BJMC -988 were used for the experiment . Extraction of enzyme : Crude Xylanase was obtained from solid state fermentation of isolates BJBC-984(2) and BJBC-988. The fermentation were carried out at 400C for four days, using wheat bran as carbon source ( with different moisture level viz 40,50,60,70 and 80% added moisture. Enzyme were extracted from the fermented bran by adding 1% NaCl solution followed by frequent stirring for 2 hours. The slurry was filtered through filter paper ( Whatman no.) . Thus the clean enzyme extract was obtained and used for further experiment. The amount of reducing sugar in culture filtrate was determined by modified DNS method. Soluble protein in the culture filtrate was estimated by the method of Lowry et al.

Achievement/Findings:

For maximum production of xylanase under solid state fermentation proper moisture content was selected. Xylanase activity was measured under different temperature for maximum enzyme activity. pH optimization of xylanase activity of BJBC-984(2) was studied. Storage stability and specific activity of xylanase enzyme were also studied. In addition to the above experiment optimization of culture conditions for the growth and production of xylanase in Czapeck media were also attempted.
