Production of Cellulolytic Enzymes and Their Degradative Effect on Jute Productions.

Abstract/ Executive Summary:

Twenty six fungal colonies were isolated and screened for cellulolytic activity. The isolates were marked as BJM – 1,BJM-2, BJM-3,BJM-4 and so on. The isolates were primarily marked as cellulolytic one on the basis of their ability to grow on CMC medium. Rest of the isolates did not grow well indicating lack of cellulolytic activity. A growth chart of the isolates on CMC medium were shown below and on the basis of their growth rate only, the well grown isolates were screened for CMCase activity through zone formations.Only 11 strains grew well in the CMC medium indicating their cellulolytic activity. Other did not grow well indicating their lack of cellulolytic activity.

Objectives:

To use of low quality jute and jute wastes for manufacturing cost effective products.

Methodology:

Samples of rotten wood were collected from Gazipur forest,saw mills of Tongi and kawranbazar and rotten jute bags from different places in and around. The samples were kept in new polythene bags and were labeled properly.The samples were inoculated in solid agar medium distributed in petri dishes. The isolated colonies of fungi were randomly picked,purified through gradual transfer from one plate to another and the pure strain maintained on the PDA slants. The fungi were inoculated in solid agar medium distributed in petri dishes containing CMC as carbon source.The enzyme activity were detected around the colonies by the appearance of zones revealed by decoloration.The plates were flooded with 10ml congo red solution.The coloration were terminated by pouring off the dye after 20 minutes and replacing with 10ml of 5 mol/L NaCl solution. After an additional 20 minutes the salt solution is discarded and CMCase activity is revealed by a pale orange zone around the colones producing CMCase. 20 g of wheat bran was taken in a 100ml conical flask and was moistened with 10 ml water.it sterilized for 15 minutes. Spore suspension of the fungal strain was poured in the sterilized wheat bran and was incubated at 350C for 3-5 days. Enzyme was extracted by adding 100 ml water to the fermented bran and was left for 2 hours with occasional shaking . The slurry was filtered through fine cloth. Filtrates obtained were stored in the refrigerator with sodium azide as preservative.

Achievement/Findings:

A number of mesophilic cellulolytic fungi were isolated and maintained on PDA slants. Out of 26 solates,11 fungal strains screened for cellulolytic activity through zone formations. Only 8 of them formed clean zone indicating their cellulolytic activity. Five out of 8 cellulolytic fungi were used for production of enzymes in wheat bran medium.
