Utilization of Wastes from Jute Industry by Fungi.

Abstract/ Executive Summary:

The fungal strains BJS – 001, BJS -002 and BJS -003 are weak producers of Cellulase and hemicellulase enzymes.

Objectives:

To upgrade the jute wastes and to get better economic return from it.

Methodology:

Three fungal strains of three different colour: green ( BJS-001) , Black ( BJS – 002) and white (BJS – 003) were isolated . All of these organisms grew well in Mendel’s medium with CMC as carbon source but produced very little cellulase and hemicellulase activity of these strains , experiments were conducted along with two identified reference organisms viz Schizophyllum commune and Scherotium rolfsii . Raw jute , jute dust and caddis were taken as substrate for liquid and solid state fermentation .The substrates were used without pre-treatment and after treatment with sodium hydroxide.Solid state fermentation medium was prepared by moistening the substrate with mendels’ basal solution.Usually shake culture fermentation in 250 ml conical flasks were carried out for up to 10 days. Enzyme assay was based upon the increase of reducing sugar following incubation of substrate with enzyme solution .The reducing sugar released was measured by the dinitrosalisylic acid method by Miller.

Achievement/Findings:

CMCase and hemicellulase producing capability of the three new fungal isolated was studied comparing with the identified reference organisms, Schizophyllum commune and Scherotium rolfsii . Compared to S. commune and S. rolfsii the new isolates were found weak in producing cellulase and hemicellulase enzymes both in solid state fermentation and shake culture fermentation with jute, caddis or dust as carbon source.
