Salinity Tolerance of Br11-saltol Recombinant Lines at Seedling Stage.

Abstract/ Executive Summary:

Thirty-three Saltol recombinant lines of BR11 genetic background were screened against salinity at seedling stage to test the effectiveness of Saltol in the BR11 genetic background, one early progeny at BC2 were isolated and advanced to F3 and these lines were screened at seedling stage to check the level of tolerance of Saltol in BR11 genetic background. Out of the 33 lines, only two (line no. 329 and 340) showed increased tolerance close to FL378. Saltol region can be defined from 10.9 Mb to 12.2 Mb in the shoot arm of chromosome 1. The genes within this region should control the seedling tolerance of pokkali. But the candidate gene for seedling tolerance is still unidentified. In this experiment, it was proved again the presence of Saltol within this region.

Objectives:

To test the effectiveness of Saltol in the BR11 genetic background, one early progeny at BC2 were isolated and advanced to F3 and these lines were screened at seedling stage to check the level of tolerance of Saltol in BR11 genetic background.

Methodology:

Thirty-three Saltol recombinant lines of BR11 genetic background were screened against salinity at seedling stage as described by Gregorio et.al 1997. Furrow-Styrofoam plate underlined with nylon net floating in a tray having Yoshida's full strength culture solution was used for growing seedlings.Fourteen days after seeding, plants were salinized at 6 ds/m. The salinity level was raised upto 12 ds/m by increasing 2 ds/m at every alternate day. Pokkali and IR29 were used as tolerant and susceptible check, respectively. One set of plants were considered as control. Standard Evaluation System (SES) scoring and survivality data were taken 21 days after salinization. Only alive plants were considered for the estimation of survival percentage.

Out of the 33 lines, only two (line no. 329 and 340) showed increased tolerance close to FL378. The average SES of these two lines were 6.16 and 5.30 with around 90% survivality. Actually all lines need to be showed similar tolerance at seedling stage. Due to unexpected result, we are interested to see the genotype composition at the Saltol region. After surveying of all 3 markers at the Saltol region we find there are several recombinations at the Saltol region, but the lines showing increased tolerance keeping the Saltol region intact.
