Genetic Improvement of the Isolated Strain for the Enhancement of Enzyme Activity for Modification of Low Grade Jute.

Abstract/ Executive Summary:

The effects of different incubation periods on the mycelial growth of S. rolfsii showed that the growth was maxium after 48hrs of incubation of pH 4.0and 5.0 but at other pH levels from 6.0 to 9.0. The growth is as manimum after 72 hours of incubation. The organisms gave some growth at all pH levels after 24 hours of incubation. this indicates that the incubation period is important to obtain maximum growth of the organism.

Objectives:

To reduce the manufacturing cost of jute products by making use of jute cutting and auxiliary materials.

Methodology:

In 1993 the effect of initial pH of S. rolfsii was reported for enzyme production . To investigate in more with the mycelial growth and the influence of pH level . The organism was cultured on medium with potato sucrose agar adjusted to different ph viz 4.0,5.0, 6.0, 7.0, 8.0 and 9.0 . The plates were maintained at room temperature. The treatments were replicated three times. The mycelial growth of the fungal colony was observed. Three isolates namely BJCK,BJCW & BJCB were collected from rotted stem containing petridish for isolation of potential microbes supplied by Mrs. Kishwar Sultana , SO, Plant Pathology Department.
