Investigation of Different Vitamins Specially Vit .a.in Different Varieties of Jute and Mesta Leaves.

Abstract/ Executive Summary:

The research work was conducted in Biochemistry Department of Bangladesh Jute Research Institute during the year of 1995. Bita -carotene (Vitamin A), moisture, fat and ash content of jute leaves (var.D-154) were estimated at different stages of the plant growth. The percentage composition of Bita-carotene in the leaves of the variety D-154 was found to be very high i.e. 13, 537 micro g/100 g.

Objectives:

To established jute and mesta leaves as rich source of vitamin A.

Methodology:

The leaves of the jute plants, variety (D-154), at different stage of its maturity were collected from the experimental plot of BJRI(Agri.) for determining its vitamin A 9B-caotene), moisture, ash and fat content. Moisture Content: It was determined from the initial and dried weight of the samples as per standard method. Ash content: Content of the leaves was determined by heating the samples in a muffle furnace at 900 degree C for 12 hours. Ash content was calculated from the weight of the ash remaining in the crucible. Fat content: Fat content of the fresh leaves was determined by refluxing the fresh green leaves with petroleum ether (40-60 Degree C B.P.) for 16 hours. Estimation of vitamin A (Bita-carotene): Green jute leaves (0.5 g) wee cut into small pieces. Then the leaves were soaked in petroleum ether and acetone mixture (4:1 v/v) and then was smashed along with glass particles. The liquid extract from the leaves was collected several times until the samples become colorless. The collected extract was evaporated to dryness in a water bath under reduced pressure. The dried sample was extracted with a little amount of petroleum ether and then it was passed through an activated alumina column.the alumina column was then eluted with petroleum ether and 2% acetone at 40-60 degree C. The eluted solvent was collected in a 100 ml volumetric flask and petroleum ether was added to make the total volume 100 ml. The optical density was measured at 420 to 450 nm in a double Beam spectrophotometer. Vitamin A (Bita-carotene) content was expressed as micro g/100g comparing the readings with the standard solution of 0.25% potassium dichromate in water.

It is observed from the table that the percentage composition of vitamin A (Bita-carotene), fat and ash content of the leaves of the variety D-154 increased gradually with the increase of the maturity of the plant. But moisture content decreased gradually with the increase of maturation. This year one variety (D-154) was studied.
