Characterization of Some Important Vegetable and Fruit Germplasm of Bangladesh Using Morphological Traits and Molecular Markets.

Abstract/ Executive Summary:

Nine experiments were conducted during 2005-06 to study the variability and genetic divergence using morphological traits and molecular markets. The results of the experiments indicated that wide variability exists among the collected germplasm of different vegetables and fruits. In case of bitter gourd, days to first male and female flowering varied significantly and ranged from 70.33 to 81.33 days and 74.00 to 85.33 days, respectively. Sex ratio also varied widely and ranged from 15.22 to 27.22 with a mean value of 20.58. Germplasm MC138 gave the highest and MC080 gave the lowest number of fruits per plant. Yield per plant of bitter gourd ranged from 0.37 kg to 2.69 kg with an average of 1.35 kg. Among 36 germplasm of muskmelon, CML28 was early for female flowering (66.00 days) and CML21 took the highest days to flowering (85.11 days) having a mean value of 75.13 days. Significant difference was observed in case of sex ratio and it ranged from 13.63 (CML03) to 29.25 (CML29) with a mean value of 23.15. The highest number of fruits per plant (7.33) was recorded in CML03 and lowest (0.89) in CML04 and CML29. Average fruit weight of muskmelon varied widely and ranged from 0.50 (CML24) to 7.25 CML140) with a mean value of 2.67 kg. The highest percentage (6.27%) of TSS of muskmelon fruit was recorded in CML28 and the lowest (3.10) in CML29. Correlation coefficients study for both bitter gourd and muskmelon indicated that yield contributing characters had the significant and positive association with yield.

Objectives:

To study the variability and genetic divergence using morphological traits and molecular markets.

Methodology:

The present study was conducted with study the variability and genetic divergence using morphological traits and molecular markets. Morphological traits: Based on morphological traits, the germplasm of bitter gourd and muskmelon were also tested for genetic divergence using D2 techniques. The different germplasm of bitter gourd collected from the same places were grouped into different clusters indicated that there is no relationship between genetic divergence and geographical distribution of the germplasm. Molecular Characterization: For molecular Characterization of the collected vegetable and fruit germplasm, isozyme and RAPD were used. The occurrence of different types of zymotypes and banding pattern indicated a wide variability in case of peroxides, glutamate oxaloacetate transaminase and malate dehydorogenase isozyme systems among the collected brinjal, bitter gourd, muskmelon, okra and lemon germplasm. In case of RAPD analysis, 84.21, 58.00, 96.00 and 72.73% polymorphisms were observed using different primers in case of okra, sweet gourd, mango and lemon germplasm, respectively. Wide range of Nei’s (1973) gene diversity and Shannon information index across all loci of okra, sweet gourd, mango and lemon indicated the existence of high level of genetic variation among the germplasm. The germplasm grouped into a cluster using morphological characters were grouped into different cluster group when isozyme system and RAPD were used as marker.

Achievement/Findings:

The study was revealed that along with morphological characterization, molecular Characterization of the vegetable and fruit germplasm (both land races and wild relatives) is necessary to avoid duplication of the germplasm for future conservation.
