Detached Tiller Test: Protocol for Measurement of Quantitative Physiological Resistance of Rice Sheath Blight (csica)

Abstract/ Executive Summary:

Variation among the eight genotypes differed from each other in context of plant architecture as well as sheath blight disease index. Apparently a positive relationship was seen with culm width and number of sheath blight lesion. Culm width was highest in BR11 followed by Tetepand IR58. In contrast, the least culm width was found in Betichikon (4.11 mm) followed by Te-Quing these two genotypes found to be associated with the lower number of sheath blight lesion. Mean lesion length also varied among the genotypes so was furthest lesion length. The least lesion length was recorded in Te-Quing while highest with IR58. Furthest lesion length was also associated with followed by Khato Chalani and. Disease severity was least in Te-Quing followed by Beti-Chikon. At least two genotypes having partial characters in this trial even under environment favorable for disease but unfavorable for host plant were identified.

Objectives:

To develop effective protocol for measurement of quantitative physiological resistance of rice sheath blight.

Methodology:

Three independent tests were conducted using tillers of the same planting dates. There were 3 replications i.e. 3 plants of a given rice genotype in each test. RCB design was followed. In each test single tiller was placed in test tube with water and considered as replication. Earthen pots were placed inside a frame made of iron and covered by polyethylene. Tetep, Jasmine 85, Teqing, Dudruchi, Betichikon, Khatho-Chalani, IR 58 and BR11 were tested. 16-days old seedlings were transplanted with 25 x 20 cm spacing using single seedling per hill. Agronomic and other management practices were followed as per BRRI recommendation. Hills for each genotype were uprooted carefully and washed thoroughly in running water from which primary and secondary tillers were separated carefully 41, 42 and 43days after transplanting to minimize root damage and used for inoculation. Each tiller was placed in a test tube filled with distilled water (10 ml) and inserted onto sand. Four virulent isolate of R. solani was isolated from freshly infected rice sheath, purified by single tip culture and anastomosed with reference isolate IMI303152. One of them that produced abundant sclerotia within 7 days of desired size was selected. Sclerotia from seven-day-old PDA culture of R. solani having 1.5 ± 0.3 mm diameter were selected for inoculation. Single sclerotia were transferred from the petri dish and inserted 1 cm below the 2nd leaf collar from the tip, on the inner side of the sheath. The inoculated tillers for each variety in test tubes were placed onto sandfill non porus earthen pot and placed thermohygro meter near leaf canopy to monitor max –min RH and temperature. The pots were covered by poly bag and plants were exposed once in a day to exchange aeration. Water spray was required due to high relative humidity maintained inside poly covering from moist sand and evapo-transpiration by the plant. The pots were placed in a glass house at 25±20^oC under ambient photoperiod. Disease assessment on number of lesions, length of each lesion (including margin) and culm, the disease spread (distance between lowest and highest lesions), lesion with thick and dark margin, status of the leaf blade just next to the inoculation site (alive or dead) and disease severity was done 7 days after inoculation.
