Development of Mass Screening Method Against Sheath Rot Disease of Rice

Abstract/ Executive Summary:

Two inoculation methods, clipping and spraying were tested along with a control (without inoculation) was used in this experiment. In each method urea was applied at three different rates. In spraying method, plants were inoculated at maximum tillering stage by spraying with the spore suspensions of the pathogen while in clipping method flag leaves were clipped using scissors with spore suspension. Before inoculation, inoculum was prepared by sheath rot causing pathogen Sarocladium oryzae, isolated from freshly harvested sheath rot infected samples. The isolates were grown on PDA and allowed 12 days for sporulation. Then the spore masses were collected and diluted into water (100 ml water/plate) and prepare spore suspension. Data on percent tiller infection and percent unfilled grain were recorded at maturity stage. Significant difference was found in clipping method compared to spraying and control @ 80 kg N/ha. Then other N-doses in both the season. But when N dose was increased none of the methods was found significantly effective in development of sheath rot disease compared to control.

Objectives:

To identify a suitable and easier method for large scale screening against sheath rot disease of rice.

Methodology:

The experiment was carried out in the BRRI farm, Gazipur during T. Aus 2010 using BR1 cultivar and T. Aman using BRRI dhan49 cultivar. Seedlings of BR1 and BRRIdhan 49 were grown on seed-bed and seedlings were transplanted (25 days of BR1 and 35 days of BRRI dhan49) in main field with spacing of 20 × 20 cm. Before planting, fertilizers: TSP and MP were applied at recommended doses for BR1 and BRRI dhan49 (BRRI, 2000) at the time of land preparation. Three levels of nitrogen fertilizers were used as 80, 120 and 160 kg/ha during T. Aus and 80, 120 and 140 kg/ha during T. Aman in the form of urea. Urea was applied as top dressing in equal 3 splits during the plant growth. Other management practices were done properly at the experimental period.
