Development of Mass Screening Method Against Sheath Rot Disease of Rice

Abstract/ Executive Summary:

Objective of this study was to identify the easy and effective screening method against sheath rot of rice. The experiment was carried out in the BRRI farm, Gazipur during T. Aman, 2008 using BR11 cultivar. Seedlings of 35 days were transplanted with 20 cm x 20 cm spacing in 2 m x 3 m unit plots. Significant difference was found in clipping method compared to spraying method and control @ 80 kg N/ha. But when N dose was increased none of the methods was found significantly effective in development of sheath rot diseases compared to control but clipping was found to be good compared to spraying method in higher nitrogen doses (100 kg N/ha and 120 kg N/ha).Significant difference was found in clipping method compared to spraying method and control @ 80 kg N/ha. Objectives:

To develop an effective screening method against sheath rot of rice

Methodology:

The experiment was carried out in the BRRI farm, Gazipur during T. Aman, 2008 using BR11 cultivar. Seedlings of 35 days old were transplanted with 20 cm x 20 cm spacing in 2 m x 3 m unit plots. During final land preparation all fertilizers except urea were applied at recommended dose. Urea was applied in three equal splits as top-dress. Two inoculation methods, clipping and spraying were tested along with a control (without inoculation) in this experiment. In each method urea was applied at three different rates as 80, 100 and 120 kg N/ha. Each treatment was replicated three times following RCB design. In spraying method, plants were inoculated at maximum tillering stage by spraying with the spore suspensions of the pathogen while in clipping method flag leaves were clipped using scissors with spore suspension. Before inoculation inoculum was prepared from sheath rot causing pathogen Sarocladium oryzae, isolated from freshly harvested sheath rot infected samples in the previous season. The isolates were grown on PDA and allowed 10 days for sporulation. Then inoculum suspension was prepared with sterilized water @ 100 ml/plate and used for inoculation. Data on percent tiller infection was recorded at maturity stage.
