Confirmation of F1 and Backcross Breeding.

Abstract/ Executive Summary:

Objective of this study was to confirmation of the original crosses (F1s) with introgression of sub 1 gene for submergence tolerance and back crossing with recurrent to generate BC1F1 population. Two donor parents for sub1 gene were selected from IRRI developed submergence tolerance advance lines. Four recurrent parents were involved to develop the original crosses (F1s). F1 seeds of these crosses were disinfected by treating with 0.1% mercuric chloride solution for one minute. Three crosses were confirmed as true F1s in comparison with their parents.These F1s were crossed with the recurrent parents and generated three BC1F1 population also. F2 of these original crosses were preserved to proceed for conventional breeding.

Methodology:

Two donor parents for sub1 gene were selected from IRRI developed submergence tolerance advance lines. Four recurrent parents were involved to develop the original crosses (F1s). F1 seeds of these crosses were disinfected by treating with 0.1% mercuric chloride solution for one minute. The treated F1 seeds along with the donor and recurrent parents were germinated in the petridishes and then seeded on the earthen pots. Thirty day old seedlings were transplanted @ single seedling with a spacing of 25 x 15 cm in the house along with respective parents.
