Seed Health Test and Pathological Study of Jute, Kenaf and Mesta Seeds for Recommendation

Abstract/ Executive Summary:

The study was conducted to investigate the occurrence of different seed borne fungal pathogens of jute, kenaf and mesta and to determine the percentage of seed borne infections in seed samples collected from different stations of Bangladesh Jute Research Institute. Germination percentage of seed samples were determined by blotter method in the laboratory of Plant Pathology Department of Bangladesh Jute Research Institute following the rules of ISTA . After 10 days of incubation, the seeds were examined under microscope for the detection of different fungal pathogens. Pathogens Macrophomina phaseolina, Colletotrichum corchori and Botryodiplodia theobromae were found limited in most of the collected seed samples. Highest parentage of Colletotrichum corchori (8%) was recorded in BJC-7370 of Chandina regional station. Maximum parentage of Fusarium (13%) was recorded in CVL-1 collected from regional station, Chandina followed by 12 % Fusrium observed in BJC–7370 collected from the same location. The highest total pathogen (24% was recorded in BJC-7370 of Chandina regional station followed by 22 % recorded in 2142 collected from Faridpur regional station of BJRI.

Objectives:

1. To study the presence of different seed borne fungal pathogen of jute, kenaf and mesta and to determine the percentage of seed borne infections in seed samples.

2. To maintain seed standard and recommend for using disease free seed by the breeders and jute growers for successful crop production.

Methodology:

The experiment was conducted in the Pathology Laboratory of Bangladesh Jute Research Institute, Dhaka during the year 2012, Twenty seven Seed samples of breeder seed, foundation seed, certified seeds and TLS of different varieties were collected from different Jute Research Regional and Sub-stations of Bangladesh Jute Research Institute. Germination percentage of seed samples were determined by blotter method in the laboratory of Plant Pathology Department of BJRI following the rules of ISTA. After 10 days of incubation, the seeds were examined under microscope for the detection of different fungal pathogens. Percent of seed borne infections were determined for recommendation before sowing seeds in the field.
