Study of Nature and Transmission of Fungal Pathogen in Jute Seed

Abstract/ Executive Summary:

Most of the fungal pathogens in jute are seed borne nature. Seeds are both vehicles and suspect of pathogens and they may carry fungal pathogen as contamination or as infections. Seed borne nature of inoculums allows the pathogen to survive in a resting host pathogen combination. The establishment and development of infection within the seedling or subsequent adult plant is the last decisive link in the process of seed transmission. Sowing of infected seeds results poor germination and causes crop damage. Maximum percentage of stem rot disease transmission from naturally infected plants was found 19% in CVL-1 variety. Maximum percentage of stem rot disease transmission from mechanically inoculated seed borne pathogen was found in CVL-1 variety (37%). Disease transmission of fungicide treated seed ranges from 0 to 8%.

Objectives:

1. To study and measure seed borne transmission of fungal pathogen in two varieties of jute.

2. To study the different disease incidence by the seed borne transmission and for the management of disease.

Methodology:

The experiment was carried out at Jute Agriculture Experiment Station, Manikgang during the year 2012. Two varieties of jute were used in the study (CVL-1 and O-9897). Three different strategies were applied in order to study and measure seed transmission of fungal disease in two varieties 1) Seeds harvested from naturally infected plants. (II) Seeds harvested from mechanically inoculated plants. (III) Seeds treated with fungicides (Provax-200) . Jute seed sample were studied in pathology laboratory of Bangladesh Jute Research Institute. to measure initial infection and identify the fungal pathogen for the presence of the desired seed borne fungal pathogen by blotter method and PDA medium ISTA (1976) in order to get suitable seed samples to be used for the study. These three types of samples were sown in randomized complete block design with three replications. Unit plot size was 3mx2m. Seeds were sown in 8/4/2012. Usual agronomic practices were done carefully. The recommended doses of fertilizers were used. Percentage of germination was recorded after 10 days of sowing. No plant protection was measured. Number of disease affected plants was recorded at 15 days age of the plants. Symptoms of disease were studied and noted. Samples of affected plant were collected and plated in PDA medium in glass Petridis in the pathology laboratory of Bangladesh Jute Research Institute to identify pathogen associated with the disease. Transmission of the major seed borne fungal pathogen of jute from seed to plant, plant to seed was observed. Disease reactions were evaluated on the basis of percentage of affected plants in each variety.

Achievement/Findings:

Disease pathogen was detected in the seed coat and in the cotyledons. Disease symptoms were associated with the infected plants. Maximum percentage of stem rot disease transmission from naturally infected plants was found 19% in CVL-1 variety. Maximum percentage of stem rot disease transmission from mechanically inoculated seed borne pathogen was found in CVL-1 variety (37%). Disease transmission of fungicide treated seed ranges from 0 to 8%. Seed should be treated with fungicide by the growers for the management of disease which will be helpful for the production of higher fiber yield.
