Evaluation of the Efficacy of Bt Transgenic Brinjal Lines Against Brinjal Shoot and Fruit Borer Field Population in Confined Greenhouse Condition

Abstract/ Executive Summary:

Efficacies of eight Bt brinjal lines (along with their non-Bt isolines) were evaluated against the field population of brinjal shoot and fruit borer (BSFB) in the confined greenhouse of Biotechnology Division of BARI, Gazipur during March-May 2011. Two methods, oviposition preference test and direct infestation methods were applied. Significant differences in percent shoot infestation were recorded between the Bt and non-Bt brinjal lines in response to oviposition of BSFB field collected population. Similar trend of results were observed in the direct infestation method where, significantly less shoot infestation were recorded in all the Bt brinjal lines than their non-Bt version in the confined greenhouse. In some Bt lines (Chaga, Uttara and Nayantara), the infestation was high especially in oviposition preference test. So, growing of resistance by BSFB field population in those Bt lines may be faster, when those will be released for commercial cultivation.

Objectives:

To determine the effectiveness of different Bt transgenic brinjal lines against BSFB populations of different regions of the country in the greenhouse condition

Methodology:

This study was conducted in the confined greenhouse of Biotechnology Division of BARI, Gazipur during March-May 2011. Two methods were applied to determine the effectiveness of Bt transgenic lines against field collected BSFB populations. Oviposition preference test: Bt transgenic plants were grown in the earthen pots. After Bt confirmation test, 35-40 days old plants were caged (one plant of Bt lines along with their non-Bt isolines and there were eight Bt brinjal lines with non-Bt version) with 32 mesh net. The plants were set randomly following CRD within the cage. One day after caging 05 pairs of BSFB adults were released randomly in the cage, so that they can lay eggs on the preferred plants. After one month of BSFB release, the nets were removed and the number of total shoot and infested shoot per plant were counted and recorded. There were three replications.Direct infestation method: Bt transgenic plants of eight brinjal lines, viz. Uttara, Kazla, Nayantara, Khotkhotia, Chaga, Singhnath, ISD 006, Dohazari as well as their non-Bt versions were grown in the earthen pots. After Bt confirmation test, 35-40 days old Bt plants and also the non-Bt plants of same ages were directly infested with larvae of BSFB (10 neonate larvae per plant). After 20 days of BSFB larvae release, the number of total shoot and infested shoot per plant were counted and recorded. There were three replications also.
