Studies for Resistant Materials Against Biotypes of Macrophomina Phaseolina Under Controlled Condition

Abstract/ Executive Summary:

Materials of 20 germplasm accessions and 2 varieties (D-154, Lalshachi) and 5 lines of Breeding Division were tested against two virulent races (MS-6 and MS-9) of Macrophomina phaseolina under controlled condition in green house following CRD. Seeds were sown in pots and at 40 days old age the plant were artificially inoculated on stem with 48 hours old culture of 2 races of pathogen. Two plants of each line were inoculted with each race in two spots on stem. Inoculated plants were kept inside the green house chamber for 48 hours where humidity was maintained at 90%. Infected lesion developed on stem were measured after 7 days of inoculatioon. Six accessions 5073, 3744, 3746, 4119, 4630, 370 and one local variety were found resistant against two biotypes of M. phaseolina. 3 accessions 4820, 4374 and 4701 showed susceptible reaction. Rest of the accessions showed moderately resistant.

Objectives:

1. To find out the sources for resistant materials against Macrophomina phaseolina

2.To help the breeders to develop the resistant varieties against Macrophomina phaseolina.

Methodology:

A total of 20 germ plasm accessions and 2 varieties (D-154, Lalshachi) and 5 lines of Breeding. were sown in the pots. three plants (30-40 days old) of each treatment was inoculated on 3 spots of each plant with 48 hours old culture of each race grown in PDA medium. CIrcular block ( 2mm diameter) was cut with a block cutter. One block was placed on each spot on stem after pricking with a needle. The pots were kept inside the humid chamber in the green house for 48hours where 90% humidity ( about 90% & temperature 30oC was maintained. After 48 hours pots were taken out of the chamber. Records were kept: 1.Necrotic lesions was measured in mm after 7-10 days of inoculation. 2.Disease reaction was assessed on the basis of lesion size & grouped as resistant (R), moderately resistant (MR ), Susceptible (S ) and Highly Susceptible (HS ). 3.Seeds from resistant and moderately resistant treatments was collected.
