In Vitro Regeneration of Potato

Abstract/ Executive Summary:

Nodal explants of 11 potato varieties (Solanum tuberosum L.) were cultured on MS medium for six weeks to evaluate the influence of MS medium on in vitro regeneration. All the varieties initiated roots and shoots within a week. BARI Alu-28 and BARI Alu-32 were found to be better than the remaining potato varieties for in vitro production of leaves (23.00 and 23.75/explant), number of shoots (6.50 and 4.75) and number of roots (4.5 and 6.25/explant), respectively. The maximum shoot length was observed in BARI Alu-8 (11.88 cm) followed by BARI Alu-31 (11.63 cm) and the minimum shoot length was obtained from BARI Alu-30 (6.50 cm). Apical and nodal explants of 11 potato varieties were cultured on liquid MS medium containing 0.1mg/l IBA for six weeks. Plantlets were developed from both explants in liquid MS medium containing growth regulators.

Objectives:

To evaluate the regeneration potential of 11 potato varieties of BARI for in vitro conservation

Methodology:

The experiment was conducted with 11 varieties of potato such as BARI Alu-7, BARI Alu-8, BARI Alu-13, BARI Alu-21, BARI Alu-24, BARI Alu-25, BARI Alu-27, BARI Alu-28, BARI Alu-30, BARI Alu-31 and BARI Alu-32 at the In Vitro Conservation Laboratory of PGRC during 2012-13. The varieties were developed at Tuber Crop Research Centre of BARI and they are commercially cultivated all over Bangladesh. The in vitro plantlets of 11 varieties were collected from TCRC on 22 August 2012. The age of the plantlets was approximately 8 weeks. The MS media was prepared. The pH of the medium was adjusted to 5.8 before autoclaving and the media were solidified with agar @ 8 g/l. The medium was autoclaved at 1.06 kg/cm² pressure and 121°C for 15 min. Shoot tip and nodal explants were prepared under Laminar Air Flow aseptically and inoculated into long test tube (25 x 150 mm) containing 15 ml of the basal MS medium. Each nodal explant having one node and two leaves was cultured in each test tube. Aluminum foil was used as closure. The explants were incubated for six weeks in a culture room at 25 ± 1°C under about 500 lux light provided by cool-white fluorescent tubes maintaining a 16/8-hr alternate light/dark cycle and 60-70% relative humidity. The experimental set up was in a completely randomized design with four replications and each replication consisted of 5 explants. Data from the experiment were subjected to ANOVA and mean separations were carried out by the LSD using the MSTAT program. After six weeks, the potato varieties were cultured on liquid MS media containing 0.1 mg/l IBA. The apical and nodal explants having 2-3 nodes were cultured on 40 ml MS media in Erlenmeyer flask and each flask containing four explants. The explants were cultured for six weeks.

Achievement/Findings:

Effect of MS media on in vitro growth and development from nodal explant of 11 potato varieties were measured. All the potato varieties initiated root and shoot within a week of culture. Variations were observed in the ability of explants to develop roots but insignificant. BARI Alu-30 (Meridian) took the highest number of days to root initiation (6 days) and the remaining varieties initiated roots within 4 to 5 days. At six weeks culturing, the maximum number of leaves were obtained from BARI Alu-32 (23.75), followed by BARI Alu-28 (23) and BARI Alu-8 (19.75) and the minimum leaves were found in BARI Alu-27 (7.75). The maximum shoot length was found in BARI Alu-8 (11.88 cm, Cardinal) followed by BARI Alu-31 (Sagitla, 11.63cm) and BARI Alu-21 (Provento, 10.75cm). The minimum shoot length was found in BARI Alu-30 (Meridian, 6.5 cm). Significantly, the highest number of shoot was exhibited in BARI Alu-28 (Lady Rosseta, 6.5) and the lowest in BARI Alu-7 (Diamant, 2.75). The number of roots produced from the varieties ranged from 2.5 (BARI Alu-27, Spirit) to 6.25 (BARI Alu-32, Quency). After six weeks of culture the potato varieties, the plantlets were looked poor development and the size of leaf was comparatively small. Therefore, to improve the healthy plantlets, all the potato varieties were cultured in liquid MS media containing 0.1mg/l IBA in Erlenmyer flask for six weeks. Apical and nodal explants of 11 potato varieties developed the plantlets but the plantlets were not up to mark for in vitro conservation.
