Isolation of Microbes from Different Natural Sources (rotten Grapes, Rotten Orange Peel, Green Ribbon Ret, Dry Ribbon Ret, Old Culture Etc,) and Studies of Their Retting Properties

Abstract/ Executive Summary:

Microbes undoubtedly play the most important role in the retting process of jute and allied fibre plants. Jute retting is performed by the joint action of water and various microbes found in water of pond, canal, ditches, air and soil. Previous result revealed that fungi could ret jute within a short period in sterile condition. This experiment was undertaken for the searching of new retting microbes to generate new scientific information in acceleration of jute retting speed and production of good quality jute and allied fibers (JAF). This experiment was conducted in the year of 2001 in the laboratory of Fibre Quality Improvement Division. In Manikgonj station five different microbes (Aspergillus sp., Mucor sp., Mycelia sterilia, Sclerotium sp. and Penicillium sp.) were isolated from three different sources (Green ribbon ret, Dry ribbon ret and Pond water) and it was found that among these strains only Aspergillus sp. isolated from green ribbons of jute within 11 days producing B+ grade fibre. On the other hand, five different fungi were isolated from five different sources (Old culture, Orange peel, Rotten grapes, Retting effluent and Pond water) in Central station, out of which three strains (Aspergillus terreus, Aspergillus sp. and Pleurotus sp.) were found moderately retter and two strains (Penicillium sp. and Mucor sp.) were non-retter.

Objectives:

To isolate new retting microbes for generating new scientific information in acceleration of jute retting speed and production of good quality of jute and allied fibers.

Methodology:

Differen strains of organisms were collected from various natural & environmental sources such as rotten grapes, rotten orange peel, green ribbon ret, dry ribbon ret, old culture, pond water etc, and were grown in potato dextrose agar, peptone agar and other selective synthetic media at room temperature using standard microbiological methods and their retting properties were assessed on jute stem (8 cm pieces) in sterile as well as non-sterile conditions in the laboratory. The jute varieties CVL-1 and O-9897 will be grown at JAES, Jagir, Manikgonj. The green jute plants were brought to Central Research Station and the collected organisms were applied on jute plants in the laboratory of Fibre Quality Improvement Division. (N.B. pH during isola potato dextrose agar).
