Isolation of Microbes from Different Natural Environmental Sources (rotten Banana Stem, Rotten Tomato, Soil from Manikgonj, Retting Effluent, Retted Jute, Citrus Fruit, Etc.) and Study of Their Retting Properties

Abstract/ Executive Summary:

Microbes particularly bacteria and fungi are important retting organisms of jute. They are abundant in water, soil and air. Previous result revealed that fungi could ret jute within a short period in sterile condition. This experiment was undertaken for the searching of new retting microbes to generate new scientific information in acceleration of jute retting speed and production of good quality jute and allied fibers (JAF). This experiment was conducted during 2003 in the laboratory of Fibre Quality Improvement Division. Some retting bacteria and fungi were isolated from different natural sources such as soil (Manikgonj), jute field (Manikgonj), retting effluent, retted jute, citrus fruit, rotten banana stem, rotten tomato and contaminated PDA medium. Just after the isolation the organisms were used in the retting purpose and their activities were studied in both green and dry ribbons of jute. Seventeen bacteria strains and eleven fungal strains were isolated from different sources and their retting activities were tested on green and dry ribbons of jute (CVL-1). In case of bacterial retting, one strain was found the best retter and retted jute in 9 days. The other five bacterial strains were found good retter and retted long jute ribbons in 10, 12, 10, 14 and 14 days respectively. Nine strains were found non retter. On the other hand, nine different fungi were isolated from seven different sources, out of which three strains were found the best retter (retting times were 7, 6 and 7 days respectively) and one strain was found good retter, two poor retter and one moderate retter and two strains were found non retter.

Objectives:

1.To isolate and identify more active retting microbes (bacteria and fungi) from our natural environmental sources and

2.To assess their retting properties on jute and allied fiber (JAF) plants

Methodology:

Various strains of microbes were collected from various natural & environmental sources such as rotten banana stem, rotten tomato, soil from Manikgonj, retting effluent, retted jute, citrus fruit, contaminated potato dextrose agar, etc. and were grown in potato dextrose agar, nutrient broth, peptone agar and other selective/ synthetic media at room temperature using standard microbiological methods and their retting properties were assessed on jute stem (3.8 cm pieces) in sterile as well as non-sterile conditions in the laboratory. The jute varieties CVL-1 and O-9897 will be grown at JAES, Jagir, Manikgonj. The green jute plants were brought to Central Research station and the collected organisms were applied on jute plants in the laboratory of Fibre Quality Improvement Division. (N.B. pH during isolation was 5.4, Temperature during isolation was 30ºC and media used for isolation was potato dextrose agar).
