Isolation of Microbes from Different Natural Environmental Sources (decomposed Orange Peel, Rotten Papaya, Soil, Rotten Jute Plant, Rotten Citrus Fruit, Retted Fibre Decompoced Paddy Straw, Etc.) and Study of Their Retting Properties

Abstract/ Executive Summary:

Microbes particularly bacteria and fungi are important retting organisms of jute. They are available in nature specially in air, water and soil. This experiment was conducted in the period of 2004 and 2005 for the searching of new retting microbes to generate new scientific information in acceleration of jute retting speed and production of good quality jute and allied fibers (JAF) in the laboratory of Fibre Quality Improvement Division. During the year some retting microbes (bacteria and fungi) were isolated from different natural sources such as soil, retted jute, retted jute stick, decomposed orange peel, dry jute plant, rotten papaya, citrus fruit, and retted fibre. The isolated microorganisms were used in the retting of jute and their activities were studied in both green and dry jute ribbons. Out of nine strains of aerobic bacteria tested, two strains of aerobic bacteria isolated from soil were found to be the best retters which retted green jute ribbons (CVL-1) in 8 and 9 days respectively and fibre grade was A+. On the other hand, five strains of aerobic bacteria isolated from retted jute and retted jute stick were found good retters, which retted jute in 10-13 days and two strains of aerobic bacteria isolated from decomposed orange peel were found non-retters. Besides, out of seven different fungi isolated from different sources, two strains were found to be the best retters, other two strains were good retters, another two strains were poor retters and one strain was found non-retter. Again, Three strains of aerobic bacteria isolated from rotten jute plants were found to be moderate retters and retted jute in 14 days. Four strains of aerobic bacteria isolated from orange peel and decomposed paddy straw were found non-retter.

Objectives:

1.To isolate more efficient retting bacteria and fungi from natural environmental sources and

2.To assess their retting properties on jute and allied fiber (JAF) plants.

Methodology:

Different strains of microbes were collected from various natural & environmental sources such as decomposed orange peel, rotten papaya, soil, rotten jute plant, rotten citrus fruit, retted fibre decompoced paddy straw, etc and were grown in potato dextrose agar, nutrient broth, peptone agar and other selective synthetic media at room temperature using standard microbiological methods and their retting properties were assessed on jute stem (3.8 cm pieces) in sterile as well as non-sterile conditions in the laboratory. The jute varieties CVL-1 and O-9897 will be grown at JAES, Jagir, Manikganj. The green jute plants were brought to Central Research Station and the collected organisms were applied on jute plants in the laboratory of Fibre Quality Improvement Division. (N.B. pH during isolation was 5.4, Temperature during isolation was 30ºC and Media used for isolation was potato dextrose agar).

Achievement/Findings:

Besides, out of seven different fungi isolated from different sources, two strains were found to be the best retters and fibre grade was A, other two strains were good retters, another two strains were poor retters and one strain was found non-retter. Again, Fibre grade produced from best retter was A*. Four strains of aerobic bacteria isolated from soil and retted jute stick were good retter and retted jute in 10-12 days. Three strains of aerobic bacteria isolated from rotten jute plants were found to be moderate retters and retted jute in 14 days. Four strains of aerobic bacteria isolated from orange peel and decomposed paddy straw were found non-retters. On the other hand, nine different fungi were isolated from five different sources in JAES, Jagir, Manikgonj, out of which three strains were found the best retters (retting times was 8 days and Fibre grade was A & B.), two strains were moderate retters and four strains were found non retters.
