Isolation of Microbes from Different Natural Sources (retted Jute, Green Ribbon Ret, Rotten Papaya, Contaminated Pda Broth, Etc.) and Study of Their Retting Properties

Abstract/ Executive Summary:

Bacteria and fungi are the important agents for retting of jute and allied fibre plants. They are widely distributed in nature i.e. in air, water and soil. The experiment was conducted in the year of 2006 for the searching of new retting microbes to generate new scientific information in acceleration of jute retting speed and production of good quality jute and allied fibers (JAF) in the laboratory of Fibre Quality Improvement Division. Among six bacterial strains were isolated from two natural sources from Rangpur station (viz. retted jute and pond water) only one strain (number 2/2006) isolated from retted jute, could ret green jute ribbons in 8 days in the laboratory condition when retted in conical flasks at 29ºC and produced good quality fibre while the remaining five strains were found non-retters. On the other hand, 10 different fungal strains were isolated from four different sources (viz. papaya, green ribbon ret, contaminated PDA broth and water) using standard microbiological method. Out of which one strain Mycelia sp. was found moderate retter capable of retting green jute ribbons in 10 days. Two strains (Mucor sp. and penicillum sp.) were poor retters as they could ret green jute ribbons in 13 and 14 days while the remaining seven strains were non-retters.

Objectives:

To isolate more efficient retting bacteria and fungi from natural environmental sources and to assess their retting properties on jute and allied fiber (JAF) plants.

Methodology:

Various strains of microbes were collected from various natural & environmental sources such as retted jute, green ribbon ret, rotten papaya, contaminated potato dextrose agar broth, etc. and were grown in potato dextrose agar, nutrient broth, peptone agar and other selective synthetic media at room temperature using standard microbiological methods and their retting properties were assessed on jute stem (3.8 cm pieces) in sterile as well as non-sterile conditions in the laboratory. The jute varieties CVL-1 and O-9897 will be grown at JAES, Jagir, Manikganj. The green jute plants were brought to Central Research Station and the collected organisms were applied on jute plants in the laboratory of Fibre Quality Improvement Division. (N.B. pH during isolation was 5.4, Temperature during isolation was 30ºC and Media used for isolation was potato dextrose agar).
