Fibre Quality Improvement Division, Bangladesh Jute Research Institute, Manik Mia Avenue, Dhaka-1207.

Bacteria and fungi are the important agents for retting of jute and allied fibre plants. They are widely distributed in nature i.e. in air, water and soil. The experiment was conducted in the year of 2007 for the searching of new retting microbes to generate new scientific information in acceleration of jute retting speed and production of good quality jute and allied fibers (JAF) in the laboratory of Fibre Quality Improvement Division. Aspergillus sp-1, Aspergillus sp-2 and sclerotium sp. isolated from green ribbon ret and found to be the good retters which retted the ribbon in 7-8 days. Aspergillus sp-3, Aspergillus sp-4, Penicillium sp-1, Trichoderma sp. and Sclerotium sp. isolated from contaminated old culture and dry ribbon ret which could ret green ribbon in 9-10 days and found moderate retters. Penicillium sp-2, Penicillium sp-3 and Zygorhynchus sp. isolated from contaminated culture (wheat bran) which retted the ribbon in 12-14 days and found poor retter. The rest of the fungus Curvularia sp. and Fusarium sp. isolated from pond water were non-retters. Out of twelve, three aerobic bacteria were isolated from moist soil, which could ret green ribbon in 6 days and found to be the good retters. Three aerobic bacteria isolated from jute ret (jak) retted the ribbon in 8-9 days and found moderate retters. Other three aerobic bacteria isolated from pond water retted jute ribbon in 15-18 days and found poor retters. The rest three aerobic bacteria isolated from contaminated PDA medium were found non-retters.

To isolate more efficient retting bacteria and fungi from natural environmental sources and to assess their retting properties on jute and allied fiber (JAF) plants.

Different strains of microbes were collected from various natural & environmental sources such as green ribbon ret, dry ribbon ret, contaminated old culture, wheat bran, moist soil and contaminated PDA and were grown in potato dextrose agar, nutrient broth, peptone agar and other selective synthetic media at room temperature using standard microbiological methods and their retting properties were assessed on jute stem (3.8 cm pieces) in sterile as well as non-sterile conditions in the laboratory. The jute varieties CVL-1 and O-9897 will be grown at JAES, Jagir, Manikganj. The green jute plants were brought to Central Research Station and the collected organisms were applied on jute plants in the laboratory of Fibre Quality Improvement Division. (N.B. pH during isolation was 5.4, Temperature during isolation was 30ºC and Media used for isolation was potato dextrose agar).

Thirteen different types of fungi were isolated from different natural sources viz. green and dry ribbon ret (jak), contaminated old culture, contaminated culture (wheat bran) and pond water and were grown in wheat bran. Then they are inoculated in the retting of green ribbon of jute (CVL-1).
