Screening of Potential Hydrolytic Microbes for Extracellular Enzymes and Determination of Their Enzymatic Activity on Jute Retting.

Abstract/ Executive Summary:

Commercial fibre is obtained from jute and allied fibre plants. Past researches revealed that some bacteria which could ret jute plant in 5-6 days and some fungi could ret jute within 8-18 days in moist condition, and was found very active retter for water scarce areas of Bangladesh. The experiment was undertaken to screen the potential hydrolytic jute retting fungus isolated from different sources specially farmers jute field for extraction of retting enzymes and determination of their enzymatic activity on jute retting purposes in the period of 2009. With a view to ret dry ribbon there are about 9 fungal organisms were isolated during the year under report. Out of which four fungi such as sclertium sp. 3, Aspergillus terreus, Mycelia sterilia and penicillium sp. were found good retter and were used in the retting of dry jute ribbons. It was observed that Mycelia sterilia retted dry ribbon at 18 days and produced ‘C’ quality fibre. Sclerotium sp. and Aspergillus terreus retted dry ribbon at 19 and 19 days and produced ‘B’ grade fibre with cream color appearance. Penicillium sp. retted dry ribbon in 20 days and off white ‘C’ grade fiber was produced.

Objectives:

To screen the potential hydrolytic jute retting fungus isolated from different sources specially farmers field for extraction of retting enzymes and determination of their enzymatic activity on jute retting purposes.

Methodology:

Various strains of fungi were isolated from jute cultivated land, jute cut stems after harvest, jute retting effluents and many other sources and was grown on PDA, nutrient broth and other selective synthetic media at about 30oc using standard microbiological methods. They were preserved in the laboratory. The enzymes collected from the isolates were applied on green jute ribbon and green jute plant in both sterilized and non-sterilized conditions in the laboratory of Fibre Quality Improvement Division. CVL-1 jute variety was grown at JAES, Jagir, Manikganj and were brought to Central Station, Dhaka. After harvesting, while green jute plants are stepped in water, microbes enter through the cut end and stomatal opening of the bark of jute plants and retting completed after some days. During the retting process, microbes are hired by the plant due to the contents of protein, carbohydrate, fats, mucilage, gummy material, waxes etc. Micro organisms grow and multiply there and release retting enzyme on the bark of the plant and ultimately the bark become loosened. Then the fibres are extracted manually.
