Screening of Potential Hydrolytic Microbes for Extracellular Enzymes and Determination of Their Enzymatic Activity on Jute Retting.

Abstract/ Executive Summary:

The retting microbes are widely distributed in nature such as bread, Onion, Jackfruit, Mango pickle, Bread, retting effluent of Faridpur and Dhaka were the major sources of retting organisms. Bacteria & fungi are the major organisms responsible for retting jute and allied fibres (JAF) plants. The experiment was undertaken to screen the potential hydrolytic jute retting fungus isolated from different sources specially farmers field for extraction of retting enzymes and determination of their enzymatic activity on jute retting purposes. It was conducted in Fibre Quality Improvement Division in the period of 2012. Previous researches revealed that there are some bacteria could ret jute within 5-6 days in water logged condition and some fungi which could ret jute within 8-13 days in moist condition was found very suitable retter for water scarce areas of Bangladesh, especially in Rangpur and Dinajpur districts. In the study, Seven saprophytic fungi were isolated from different sources and their retting capacity was determined. It was observed that hydrolytic activity was higher in Aspergillus niger, Aspergillus sp. and A. terreus sp. retted green jute cut at 10 days, 12 days and 10 days respectively. Those fungi produced cream, Light Cream fibre respectively. The fungi Sclerotium sp. and penicillium sp. retted green ribbons at 13 & 14 days respectively. Rhizopus sp. retted green ribbon at 15 days and the rest two fungi were found non retter.

Objectives:

To screen the potential hydrolytic jute retting fungus isolated from different sources specially farmers field for extraction of retting enzymes and determination of their enzymatic activity on jute retting purposes.

Methodology:

Various strains of mixrobes (bacteria and fungi) were isolated from jute cultivated lands, the jute cut stems after harvest, jute retting effluents and many other sources and was grown on PDA, nutrient broth and other selective synthetic media at about 30oc using standard microbiological methods and they were preserved in the laboratory. The enzymes which were collected from the isolates will be applied on green jute ribbon and green jute plant in both sterilized and non-sterilized conditions in the laboratory of FQID. The jute variety Bjri tossa pat-4 were grown at JAES, Jagir, Manikganj and wiere brought to Central Station, Dhaka.
