Preparation of Fungal Inoculum Package with Promising Fungus to Be Used in Jute Retting in Water Scarce Jute Growing Areas.

Abstract/ Executive Summary:

At present condition in Bangladesh, jute retting is a severe problem due to acute shortage of retting water. Fungal retting has a great impact for water scarce areas like Rangpur, Dinajpur and Bogra etc. The experiment was undertaken to develop an appropriate fungal inoculum package and to ascertain the viability and storing capacity of promising jute retting microbes grown in different medium/ carrier materials. It was conducted in Fibre Quality Improvement Division in the period of 2012. It was observed from the perevious study that wheat bran medium could support the growth of the fungi better than soil, rice husk and beggase. So wheat bran was selected for the preparation of inoculum package. From the results, it was also observed, after 4-5 days fungal growth innoculum. At 7 days and 10 days inoculum was used in 2 set of experiment. It was observed that 7 days inoculum was better active than 10 days inoculum.

Objectives:

To develop an appropriate fungal inoculum package and to ascertain the viability and storing capacity of promising jute retting microbes grown in different medium/ carrier materials.

Methodology:

Jute plants (Bjri tossa jute-4) were grown in JAES, Jagir, Manikganj and Central Station, Dhaka. Plants were ribboned and were dried. The promising fungus was inoculated in sterilized wheat bran, baggase, saw dust and sandy loam soil separately for their growth and multiplication. Then the package was made. Viability of the fungal inoculum was tested. Fungal inoculum was spreaded on the green jute ribbons and it was covered with moist Hessian. Day to day keen observation was kept on the progress of retting. Produced fibres were assessed in respect of quality. Viability and storage capacity of the promising microbes grown in saw dust and other media were assessed. 100 g wheat bran was taken in 12 conical flask with 50ml tap water and was mixed well. The water soaked wheat bran was kept in 500 ml conical flask and was cotton pluged. After autoclave the indigenous wheat bran medium was inoculated with 6 fungi and was kept in growth chamber taken in 12 conical flask including 2 control flask.
