Genetic Analysis and Molecular Characterization of F1 Population of BINAdhan – 7 × Fl - 378

Abstract/ Executive Summary:

To introgress salt tolerant rice gene, a cross was made between Binadhan-7 (short duration HYV) and FL-378 (exotic salt tolerant rice line) where Binadhan-7 was recipient parent and FL-378 was donor parent. One hundred and thirty four F₁ seeds were developed. F₁ plants were grown to evaluate phenotypical performance and for further backcrossing. In F₁population, average plant height, average number of productive tillers per plant, days to flowering and days to maturity were 85.61 cm, 33.79, 93.1 days and 124.6 days, respectively. Significant negative heterosis was observed in plant height, days to flowering and days to maturity. Significant positive heterosis was observed in number of productive tillers per plant. Out of 8 SSR markers, RM510, RM407 and RM18 showed distinct polymorphism and were selected to confirm the hybridity status of the F₁ population. RM407 and RM18 identified 17 plants as heterozygous. But RM510 identified 15 heterozygous plants. These heterozygous plants can be used for further backcrossing. The good responded markers can be efficiently used in tagging salt tolerant genes and in marker assisted selection.

Objectives:

To introgress saltol gene from FL-378 to Binadhan-7 by crossing method and 2) to evaluate the phenotypic performance and molecular characterization of F₁population.

Methodology:

The experiment was carried out in experimental farm yard and laboratory of Biotechnology Division, Bangladesh Institute of Nuclear Agriculture (BINA), Mymensingh from July, 2011 to October, 2012. On the basis of previous result two rice genotypes viz., Binadhan-7 (HYV and short duration) and FL-378 (exotic salt tolerant line) were selected as parents for transferring salt tolerant genes (Islam, 2004) from tolerant line to high yielding rice variety. The parental lines were crossed in Aman season, 2011 to develop F₁ seeds where Binadhan-7 was the recipient parent and FL-378 was the donor parent. One hundred and thirty four seeds were collected and sown in Boro season, 2012 to develop F₁ plants. Only 64 plants survived. Data were collected on plant height, number of tillers per plant, number of effective tillers per plant, days to 50% flowering and days to maturity. Relative Heterosis, heterobeltiosis, gene and genotypic frequency were estimated by statistical analyses. Young vigorously growing fresh leaf samples were collected from 21-day old seedlings to extract genomic DNA. DNA was extracted from leaf samples following CTAB method. Eight primers of random sequence were used for parental survey. Among them three polymorphic SSR markers viz., RM407, RM18 and RM510 were selected to evaluate F₁plants for hybridity test.

The heterozygous F₁ plants can be used for further backcrossing to introgress saltol gene into Binadhan-7 and the tested markers RM407, RM18 and RM510 can be used in backcrossed population to select the introgressed salt tolerant rice line.
