Screening of White Jute Accessions Through Anatomical Studies for Higher Fibre Content.

Abstract/ Executive Summary:

In this experiment, fiber bearing potentiality of 24 advanced lines along with four released varieties BJRI Deshi Pat-7, BJRI Deshi Pat-8, CC-45 and CVL-1 were assessed through anatomical characters such as bark thickness, number of phloem pyramid (trapezoid), area of phloem pyramid, number of layer, number of fiber bundle. Significance differences were observed among the treatments for number of pyramid and number of layers. Advanced line C-12221 has higher fibre bearing potentiality followed by C-5036, C-5044 and C-5149

Objectives:

1.To find out bark thickness of the stem of bast fiber crop.

2.To isolate higher fiber bearing potentiality genotypes through anatomical studies

3.To find fiber compactness per unit area of plant bast.

Methodology:

5-10 plants having similar height and base diameter were considered as a sample. For studying anatomical characters 3 inches long piece of stem was cut from the middle parts of each of the selected plant at the age of 110 days. These materials were preserved in Formalin-Aceto-Alcohol (FAA) solution ( 10 ml of Formalin, 5ml of 85% glacial acetic acid and 85 ml of 70% alcohol). The preserved sample were taken out and washed. About 20-25µm thick transverse sections (TS) was done with the help of a microtome machine for preparing slide. The slides were studied under compound microscope (4X10 magnifications) and data were recorded for bark thickness, number and area of phloem wedges, number of fibre bundle layers and number of fibre bundles per section. Plant weight, base diameter and dry fibre weight of the materials were also recorded.Meceration of fibre cell: The fibre were extracted following the method described by Miah (1964) with little modification. The anatomical data were recorded from 5-10 randomly selected plants of each genotype from each replication at the time of harvest (110 days of crop age). Data were recorded on date of sowing, germination, 1^st and average flowering. bark diameter, bark thickness, number of trapezoids/section were calculated for three plants of each accession under microscope. Area of trapezoid/section (μm): Area of trapezoid was measured by the following formula: (Area of trapezoid = ½ (Upper length + Lower length) X perpendicular length of trapezoid X conversion factor [Conversion factor = 39.4 in 4 X 10 magnification], number of layers/ trapezoid, number of bundles/trapezoid were calculated under microscope from randomly selected trapezoid. Length, breadth and length: breadth ratio to be measured for 10 randomly selected fibre cells by using oculometer under microscope.

Fibre bearing potentiality of 24 advanced lines along with four released varieties BJRI Deshi Pat-7, BJRI Deshi Pat-8, CC-45 and CVL-1 were assessed through anatomical characters such as bark thickness, number of phloem pyramid (trapeziod), area of phloem pyramid, number of layer, number of fibre bundle. Seeds were sown on 04.04.2013 at Manikganj in a RCBD design with three replications was followed. Significance differences were observed among the treatment for number of pyramid and number of layers. From the data it is revealed that advanced line C-12221 has higher fibre bearing potentiality followed by C-5036, C-5044 and C-5149
