Effect of Commercial Probiotics ( Bacillus Sp) on Brackishwater Shrimp

Abstract/ Executive Summary:

Commercial probiotics was applied in nine ponds, each of 0.1ha having different stocking densities (6, 8, 10 PL/m²) of black tiger shrimp for a culture period of 120 days. Water qualities of the treatment ponds stayed well in comparison to control ponds. Average body weight and total harvest achieved were also better in the treatment ponds than the control ones. Quantitative load of total heterotrophic bacteria (THB) increased whereas the quantity of Vibrio sp. Decreased in the treatment ponds. As a result, ratio of THB to Vibrio sp. Also increased sharply both in the pond water and sediment after application of probiotics. But the quantitative profile of similar bacteria in the control ponds remained in the trend of increasing that were observed in the treatment ponds before application of probiotics.

Objectives:

To examine the effects of Bacillus probionts on microflora, water quality, survival and growth of the shrimp, Penaeus monodon.

Methodology:

The experiment was conducted in Brackishwater Station of Bangladesh fisheries Research Institute. A total of nine ponds each having 0.1 ha area was selected in station’s pond complex ofr the study. Shrimp post larvae were reared in earthen ponds and probiotics was added in monthly interval only in farming stage during 5^th March to 4^th July, 2007 of the experimental period. A three-treatment experiment in duplicate among six ponds having a set of controlled pond for each treatment was designed. Probiotics were soaked in 4 times of water and kept overnight at room temperature for germination. The rehydrated probiotics was added directly to the water at the recommended dose of 5 ppm in three different times. P. monodon post larvae were collected from a commercial hatchery (PCR teasted for White Spot Syndrome Virus). The PL15 were stocked in the in-pond nurseries under experiment. After 15 days, nurseries were opened to let the nursed shrimp graze in grow out site. THB and Vibrio sp. Were monitored bi-weekly from the water and sediment. Water samples were sieved and then 1 ml of water sample was taken for decimal dilution. Counting of microbial load from the sediment was also preformed following the similar method of dilution but sediment samples were taken as 1 g. The number of colonies in appropriate plate (20-200 clolnies in 90 mm plate) was counted after incubation for 48 hrs at 26⁰C. A randomized complete block design (RCBD) was used in each of the three experiments. Normality of the data was tested using Anderson-Darling test (MINITAB software). A significance level of P<0.05 was used for all tests.

The study might have led to enhanced digestion and increased absorption of food, which in turn contributed to the improved SGR.
