Microtuber Production of Six Potato Varieties as Affected by Medium and Photoperiod

Abstract/ Executive Summary:

The effect of medium and photoperiod on microtuber induction of some exotic and indigenous potato varieties were studied. MS medium with two thirds concentration of nitrogen and potassium salts incubated in total darkness was found most suitable for microtuber induction. The performance of the exotic varieties was much better than the indigenous ones.

Objectives:

To study the effect of some important factors like medium and photoperiod on in vitro microtuber production of some indigenous and exotic potato varieties of this country.

Methodology:

The experiment was carried out in the Tissue Culture Laboratory of IPSA during the period from July 1993 to April 1994. The virus free in vitro produced potato plantlets of four indigenous (Ausha, Challisha, Lal Pakri and Sada Gutti) and two exotic (Kufri Sindhuri and Cardinal) varieties were used for this experiment. The effect of medium and photoperiod at following different levels were studied .

Achievement/Findings:

These results give a probable indication that with exposure to longer photoperiods the endogenous stimulus present in tissue cultured single-node cutting is metabolized or formed that affected the induction of microtuber. The inhibitor synthesized under increasing photoperiods could very well be a gibberellin-like substance , as proposed by several authors (Okazawa and Chapman , 1962; and Hammes and Beyers, 1973).
