Effect of Month of Collection on Survivability and Growth of Guava Explants on a Tissue Culture Medium

Abstract/ Executive Summary:

A tissue culture experiment in nine different months, viz., August, September, October, November, December, January, February, March and April with three varieties of guava was carried out at the Plant Breeding and Gene Engineering Laboratory, Department of Botany, University of Rajshahi, Bangladesh during period August 2005 to July 2007. The objective was to study the effect of month of tissue culture on the response, survivability and contamination rate of guava explants. After one month of inoculation, lowest percent (10%) of contamination was found in BARI guava-1 in the month of February. Minimum days (7) were required to respond in the month of February. When BARI guava-1 was inoculated in the month of February, number and percentage of explants responded, number and percentage of explants survived was highest, which was followed by March and April. Growth parameters such as number and length of shoots and number of leaves were high in the month of February, followed by March and April.

Objectives:

1. To find out the effect of month of collection on survivability and contamination rate of guava explants.

2. To select suitable month guava explants collection for tissue culture.

Methodology:

The present investigation was carried out at the Plant Breeding and Gene Engineering Laboratory, Department of Botany, University of Rajshahi, Bangladesh during the period August 2005 to July 2007. Guava explants were collected in the month of August, September, October, November, December, January, February, March and April from young and mature trees of three guava varieties namely BARI guava-1 (V₁), BARI guava-2 (V₂) and Kashi (V₃). Both seedlings and mature plants were grown at Fruit Research Station, Binodpur, Rajshahi. In the present investigation mercuric chloride (H_gCI₂) and ethanol were used as sterilizing agent. Savlon was used as detergent. Murashige and Skoog (1962) medium (MS medium) was used in the experiment. Before inoculation, the collected explants were kept in front of the running laminar air flow cabinet for 30-40 minutes to reduce the chance of contamination and this was done according to Fitchet (1989). After inoculation to control browning, after about one week the brown tissues on the explants were removed and the explants were transferred to a similar fresh medium and it was repeated every week for about four weeks to minimize further browning of the tissues. The temperature of cultures was maintained at 27⁰C under the warm while fluorescent tubular lamps intensity, which varied from 2000-3000 lux. The photoperiod was maintained generally with 16 hours light and 8 hours dark. During the growth of shoots, always 16 hours photoperiod was maintained. The experiment was arranged in Completely Randomized Design with three replications. Each unit in the bottle was considered a replication. Data were collected on percentage of contamination, days required to respond, number and percentage of explants respond, number and percentage of survived, number and length of shoots and number of leaves. The collected data on different parameters were statistically analyzed. The differences among treatment means were compared using standard error Least Significant Difference (LSD) test.

Achievement/Findings:

From the above observations and discussions it may be concluded that February was the suitable month for guava tissue culture and BARI guava-1 showed the best performance on almost all parameters.
