Genetic Variation in Eggplant Cultivars Thought Rapd Market and Its Relationship to Collar Rot Disease

Abstract/ Executive Summary:

Random amplified polymorphic DNA (RAPD) technique was used for assessing genetic variation and relationship among twelve varieties of eggplant. The experimental populations were also evaluated for reaction to collar rot disease. Inoculation of eggplant varieties was done with Sclerotium rolfsii under confined field condition. Among twelve eggplant varieties, IBM 9 and Chega were found resistant and moderately susceptible and other varieties were susceptible r=to S. rolfsii. Amplification with 3 decamer primers produced 19 RADP markets of which 10 (52.63%) were polymorphic. Loci and 9 (47.37%) were monomorphic. The Unwighted Pair Group Method of Arithmetic Means dendrogram constructed from Nei’s genetic distance produced two main Eleven varieties were in cluster 1 while only one variety IPM 9 was in the cluster 2. Comparatively higher genetic distance (.3127) and lowest genetic identity (.7314) were recorded for IPM 9 vs. Mirersharai combinations. Genetic variation and relatedness to disease reaction as assessed using PARD markets could be potential sources for breeding program for the development of collar rot resistant variety which is the serious biotic stresses in eggplant and many other crops.

Objectives:

To determine the genetic variation in eggplant cultivars through RAPD and the relation of their reaction to collar rot disease.

Methodology:

Twelve varieties of eggplant namely Chega, Laffa S, Dohazari G, Jessore L, Ishurdi L, ISD 006, IPM 9, Marichbegun S, Marichbegun L, IPM 31, Singnath and Mirersharai were used in the study. The experiment was carried out in the field laboratory of Dept. of Plant pathology, BAU, Mymensingh in RCBD with 3 replications. The plot size was 2.5 m x 4.0 m maintaining row to row and plant to plant distances 75 cm. fertilizers were applied in recommended dose. Irrigation and intercultural operations were done when necessary.Inocula of the pathogen were prepared through barley culture method. Inoculation was done at early flowering stage in the afternoon. Six plants were individually inoculated by mixing 10 g of infested barely grain at plant base and covered with moist cotton. Mortality was recorded at an interval of 5 days up to 25 days and expended as percentage. Finally data were analyzed on computer based program me MSTAT. The experiment was carried out in the Biotechnology Laboratory of Bangladesh Institute Nuclear Agriculture (BINA), Mymensingh. Fresh leaf samples from 14 days old seedling were used of DNA extraction. Modified CTAB mini-prep method was followed electrophoresis on a .8% agarose gel. Single arbitrary primers were tested for their ability to amplify scorable and reproducible DNA fragments. Primers resulting in faint or unreproducible bands were excluded from subsequent analysis. Finally three primers were selected for this study.

Achievement/Findings:

The result of the present study shows diversity among the 12 varieties of eggplant which will be helpful to undertake program for the development of Sclerotium rolfsii resistant variety or any crop improvement program for eggplant through Biotechnology approaches. However, future research with additional eggplant varieties with more number of primers is needed to make conclusive remarks about genetic variation and relatedness which will enrich the gene pool.
