Callus Induction and in Vitro Regeneration of Gladiolus (gladiolus Grandiflorus)

Abstract/ Executive Summary:

The experiment was carried out during the period from July 2008 to March 2009 at the Biotechnology Laboratory, Department of Biotechnology, BAU, Mymensingh. For this purpose leaf explants was chosen. The combine effects of different concentrations of NAA (0, 1.5, 3.0, 6 mg/l) and BAP (0, 1.5, 3.0, 6.0 mg/l) on Callus induction were investigated. The highest percent of Callus were 94.44% in the Variety GL4 and lowest percent of callus were 5.55% in Variety GL10. The calli derived from leaf discs were cultured on shoot induction MS medium with BAP (0, 1.5, 3.0, 6.0 mg/l) and Kn (.5, 1.0, 1.5, 2.0 mg/l). Shoot multiplication in successive sub-culture was also possible. The highest parentage of shoot regeneration was 91.66% and highest shoot length was 4.24 cm with a minimum number of days (13) was observed in the MS medium with 3.0 mg/L BAP and .5 mg/L Kn application. Rooting of regerant shoot grown shoot was achieved on MS medium supplemented with 1.0 mg/l IBA and 1.5 mg/l GA₃ application.

Objectives:

To study the regeneration potential of four Gladiolus varieties and
to develop a protocol from leaf disc of Gladiolus.

Methodology:

The experiment was carried out during the period from July 2008 to March 2009 at the Biotechnology Laboratory, Department of Biotechnology, BAU, Mymensingh.the planting material were collected from Germplasm center, BARI, Gazipur. MS medium containing standard salts and vitamins, 3% (w/v) sucrose and .7% (w/v) agar were used. The P^H of the medium was adjusted to 5.8 before adding agar. The medium was autoclaved at 1.06 Kg/cm² for 20 min. at 121⁰C. Culture were incubated at 10000 lux with cool white fluorescent light and maintained at 24± 2⁰C. callus was initiated from fifteenth days old sprouts of leaflets cultured In vitro. Each sprout was sliced into 2 mm thick transverse sections and they were placed with a cut surface in sucros, .7% agar (BDH) and the following in 1.0 mg/L: glycin, 2.0; thiamine, 1.0, pyridoxine, .5, nicotinic acid, .5; menositol, 1000.0 and various hormonal combinations. Sterilized leaves were cut into strips avoiding the midrib and placed side down onto the media with the different concentrations of growth regulator NAA and BAP (0, 1.5, 3.0, 6.0 mg/l) and Kn (.5, 1.0, 1.5, 2.0 mg/l) for shoot initiation, they sub cultural and MS medium supplemented with combination of GA₃ (.5, 1.0, 1.5, 2.0 mg/l) and IBA (.5, 1.0, 2.0 mg/l) in order to allow root formation.

Achievement/Findings:

In this experiment, an efficient and reproducible protocol for the plant let regeneration from leaf discs explants in Gladiolus was developed. This protocol has the potential for the In vitro multiplication and improvement of Gladiolus. The protocol will be also be promising for propagation of disease free Gladiolus Spp cultivars and screening of somaclonal variants. More over, it also provides with a source of cells for genetic transformation.
